An extended developmental study of ?-glutamyltranspeptidase in rat liver plasma membranes: identification of specific patterns of changes in activity in the adult as well as the neonatal state

Abstract: Homogenates and plasma membranes were isolated from the livers of male Fischer 344 rats ranging in age from 19 hr to 92 days postnatal. These plasma membranes exhibited comparable levels of purity: protein yields were 2-2.5%; relative specific activities of 5'-nucleotidase and ouabain-sensitive Na+/K(+)-ATPase were from 8-11 and from 12-19, respectively. 5'-nucleotidase and ouabain-sensitive Na+ K(+)-ATPase displayed distinct and different developmental patterns. The activity of gamma-glutamyltranspeptidase wa… Show more

“…In accord with our previous findings [7,19,20], we show that this enzyme is expressed at low levels in the liver of rats fed the choline sufficient control diet and also that it is higher in females than in males. The response to the hepatocarcinogen diethylnitrosamine was found to be minimal in those rats on the choline sufficient control diet but markedly elevated in those on the choline deficient diet, coincident with the appearance of the hyperplastic premalignant nodules.…”

“…Following decapitation, blood was collected from the trunk of the rat , as we have done in the past [19][20][21][22], in chilled 50 ml beakers containing 70 µl of an anticoagulant solution containing 10.5 mg K + EDTA and 0.014 mg potassium sorbate. The whole blood was then centrifuged at 2000g at 4°C for 10 minutes.…”

“…In order to achieve these objectives, we chose the Fischer 344 rat, whose development we have studied 23,24 and exposure of these rats to either regular rodent chow or 10% flax chow from before birth until the 86th postnatal day. Ten per cent flax was chosen since reports in the literature ascribe beneficial results using this flax proportion.…”
