An improved DNA method to unambiguously detect small hive beetle Aethina tumida, an invasive pest of honeybee colonies

Abstract: The scavenger and invasive species Aethina tumida threatening the honey bee has been recently introduced in Europe. We present a new, reliable and rapid multiplex real‐time PCR for efficient diagnostics enabling surveillance programs. © 2018 The Authors. “…The real-time PCR primers and probes tested were from Silacci et al 7 and Ward et al 8 ( Table 1). The SHB assay from Ward et al 8 was used in conjunction with an invertebrate control assay from Silacci et al 7 Real-time PCR reactions were comprised of 1 μL DNA in 10 μL reactions containing 5 μL iTaq™ Universal probes reaction mix (BioRad, Hercules, CA, USA), 375 nmol L -1 of each primer and 125 nmol L -1 probe for the Aethina tumida assay and 200 nmol L -1 of each primer and 50 nmol L -1 probe for the control assay. Reactions were carried out on a ViiA™ 7 real-time PCR system (ThermoFisher Scientific) using the following cycling conditions: 95°C for 1 min followed by 40 cycles of 95°C for 15 s and 60°C for 60 s. Each run included no template (negative) and Aethina tumida (positive) controls.…”
