An Injectable, Bifunctional Hydrogel with Photothermal Effects for Tumor Therapy and Bone Regeneration

Abstract: Bone Tumor TherapySignificant attention has been focused on bone tumor therapy recently. At present, the treatment in clinic typically requires surgical intervention. However, a few tumor cells remain around bone defects after surgery and subsequently proliferate within several days. Thus, fabrication of biomaterials with dual functions of tumor therapy and bone regeneration is significant. Herein, the injectable hydrogel containing cisplatin (DDP) and polydopamine-decorated nano-hydroxyapatite is prepared via… Show more

“…For example, Wang's groups have designed alendronate (ALN)-anchored PDA nanoparticles to release the anticancer drug, 7-ethyl-10-hydroxycamptothecin 80 . Similarly, injectable chitosan hydrogels containing PDA-decorated hydroxyapatite (HAp) nanoparticles could realize localized controlled and sustained release of cisplatin when exposed to NIR light 81 . Moreover, a kind of dual responsive drug delivery system, which could be triggered by both NIR laser irradiation and acidic stimulus, has been developed for bone cancer therapy.…”

“…Moreover, the promoted rBMSCs proliferation, adhesion, and differentiation in vitro as well as elevated mineral deposition in rat cranial defect in vivo also verified the bone regeneration effect of this scaffold. Another example was OSA‐CS‐PHA‐DDP hydrogel synthesized by Shiyu Luo's group 39 . PHA was composed of polydopamine (PDA) and n‐HA, in which PDA offered abundant functional groups 194,195 for cell adhesion and migration.…”

“…Hydrogel is adequately qualified for this job due to its specific sensibility to different signals, controlled drug release capacity, and facilitating histogenesis function, so once the abnormal tissue has been eliminated, surrounding normal host stem cells can directionally move to the defect area and then form new tissues through proliferation and differentiation 36 . Several attempts based on this idea have been successfully realized 37‐39 . Moreover, with a tendency to accurate and individualized therapy in the future, hydrogel is expected to be a promising option in combined treatment of localized tumor therapy and tissue engineering.…”

“…Therefore, PDA‐containing materials have been considered to have great potential in the field of tumour therapy [77]. Luo et al [59] prepared an injectable hydrogel containing cisplatin (DDP) and PDA‐modified nano‐hydroxyapatite through a Schiff‐base reaction between the aldehyde group on oxidised sodium ALG and the amino group on CS (Fig. 8).…”
