ABSTRAKFukoidan merupakan polisakarida sulfat yang berasal dari rumput laut cokelat. Salah satu faktor yang mempengaruhi mutu fukoidan adalah metode ekstraksi. Pada penelitian ini dilakukan optimasi ekstraksi fukoidan kasar dari rumput laut Sargassum binder Sonder untuk mendapatkan rendemen dan mutu paling tinggi. Optimasi tersebut dilakukan dengan empat metode ekstraksi yaitu: HCl 0,1 N pH 4 pada suhu kamar selama 6 jam, HCl 0,1 N pH 4 pada suhu 85 °C selama 4 jam, akuades pada suhu 85 °C selama 4 jam dan larutan CaCl 2 2% pada suhu 85 °C selama 4 jam. Untuk masing-masing metode dilakukan 3 kali ulangan dengan bahan baku yang sama dan pada waktu berbeda. Parameter mutu fukoidan yang dianalisis meliputi: rendemen, total gula, asam uronat, kadar sulfat dan gugus fungsi polisakarida sulfat dengan FT-IR. Rendemen tertinggi diperoleh dengan metode ekstraksi HCl 0,1 N pH 4 pada suhu 85 o C selama 4 jam sebesar 6,0% dan terendah dengan metode ekstraksi menggunakan larutan CaCl 2 2% sebesar 2,57%. Kandungan sulfat tertinggi diperoleh dengan metode larutan CaCl 2 2% selama 4 jam sebesar 8,69%. Metode ekstraksi kasar fukoidan dari S. binderi Sonder yang paling optimum menggunakan pelarut air pada suhu 85 °C selama 4 jam dengan rendemen dan kandungan sulfat yaitu 3,36% dan 8,10%. Proliferation of the introduced brown macro alga Sargassum muticum is known as a natural and hard to control phenomenon occurring along the Atlantic coasts. The phenomenon causes serious troubles for local ecosystems including the alteration of ecosystem structure, the reduction in indigenous biodiversity and economic losses (tourism, aquaculture). However, despite the serious troubles caused by S. muticum, this species contains highly remarkable bioactive metabolites. This macro alga is at present under-exploited and the valorization of its metabolites to give a positive value to this seaweed could be a solution of ecosystemic service. Biorefinery process could be one solution to valorize S. muticum. Comprehensive knowledge concerning the biochemical composition of S. muticum and the impact of environmental factors, particularly seasons, on its composition is a prerequisite before its valorization. In this study, the biochemical composition of S. muticum was evaluated by using classic colorimetric methods based on chemical analysis and Fourier Transform Infra-Red spectrometry (FTIR), and was used as a rapid and safe method that could bring advantages in screening studies and a more comprehensive management and use of seaweed products. Our results are globally in accordance, notably for phenolic compounds, showing the relevance of the use of infrared spectrometry. Moreover, based on the absorption bands of some specific and valuable compounds shown by FTIR, there was a seasonal variation in the polysaccharides, i.e., uronic acids and sulphated compounds, together with phenolic contents of S. muticum.

Extracted and depolymerized alginates from brown algae Sargassum vulgare of Lebanese origin: chemical, rheological, and antioxidant properties

International audiencePurified sodium alginate (PS alginate) was isolated from the brown seaweed Sargassum vulgare collected from the Lebanese Mediterranean coast and then depolymerized into homopolymeric polyguluronate (PolyG) and polymannuronate (PolyM) blocks by controlled acid hydrolysis. These fractions of PS alginate issued from S. vulgare were characterized in terms of composition and structure by SEC, elemental analysis, FTIR and 1H and 13C NMR spectroscopy. An alginate with a low content of protein (\textless0.62 %) and a molecular weight of 110 200 g mol-1 was identified as sole polysaccharide. Depolymerized PS alginate fractions, PolyG (32.6 %) and PolyM (22.3 %), were found to have close molecular weights, of 7500 and 6900 g mol-1, respectively. From NMR analysis, values of F G, F M, M/G ratio, F GG, F MM, and F GM (or F MG) blocks were determined and compared with those of alginates from S. vulgare of Brazilian origin and other Sargassum species. Our PS alginate appeared different from the Brazilian S. vulgare alginate, with a lower M/G ratio (0.785 instead of 1.27), a predominance of the G blocks (F G and F GG \textgreater 0.5) instead of the M blocks, and it showed more similarity to the composition of some alginates extracted from other species of Sargassum. High G or M contents (≥80 %) were measured from PolyG and PolyM blocks, respectively. The viscosity of the PS alginate and its fractions PolyG and PolyM was determined. PS alginate from S. vulgare of Lebanese origin showed a Newtonian flow behavior for concentration lower than 0.5 % in 0.1 M NaCl solution, while a shear-thinning pseudoplastic behavior is observed for concentration range between 0.75 and 10 %. Also, storage (G′) and loss (G″) moduli were studied for two concentrations of PS alginate solutions (5 and 10 %). Antioxidant properties of the non-depolymerized and depolymerized alginates were evaluated by determining the scavenging ability of the stable radical DPPH (2,2-diphenyl-1 picrylhydrazyl). Clearly, the results demonstrated differences in radical scavenging efficacy between PolyG and PolyM fractions. The higher hydroxyl radical scavenging activity was observed for the PolyG fractions (~92 % at 2 mg mL-1) and this activity was comparable with those of standard antioxidants. Abstract: Phaeophyceae, Ochrophyta) are analyzed by spectroscopic techniques. The nature of the polysaccharides (with extraction and without any type of extraction) present in these seaweeds was determined with FTIR-ATR and FT-Raman analysis of extracted phycocolloids and ground dry seaweed.
