Analysis of Charged Cyclodextrin Derivatives by On-Line Capillary Electrophoresis-Ionspray-Mass Spectrometry

Abstract: On-line capillary electrophoresis-ionspray-mass spectrometry (CE-ISP-MS) was applied to a composition analysis of charged cyclodextrin (CD) derivatives. Quaternary ammonium b-CD (QA-b-CD) and carboxymethylated g-CD (CM-g-CD), which were used as chiral selectors for enantiomer separations by CE, were analyzed. As for QA-b-CD, eight components having different degrees of the quaternary ammonium substitution could be identified based on their mass spectra. The components were detected as either molecular ions or … Show more

“…Table 1 (see Addendum) reports some selected examples of such application. Typically, carbohydrates carrying ionizable groups, like carrageenan oligosaccharides [51], or carboxymethylated and quaternary ammonium-CDs [47] are not derivatized. Neutral saccharides, on the contrary, are normally linked to negatively charged tags like p-amino benzoic acid (p-ABA) [42], 8-aminonaphthalene-1,3,6-trisulfonic acid (ANTS) [48], APTS [49], 7-amino-1,3-naphthalene disulfonic acid (ANDSA) [50] or, more rarely, to positively charged molecules, like 3-(acetylamino)-6-aminoacridone (AA-Ac) [52].…”

“…To date, the main articles regarding the CE-MS of saccharidic compounds found in literature include the analysis of (i) standard mixtures [42,[47][48][49][50][51][52]; (ii) carbohydrates in nutrition [19,[53][54][55][56][57][58][59]; (iii) glycans from glycoproteins [60][61][62][63][64][65]; (iv) carbohydrates in drug development [45,[66][67][68][69][70][71][72][73][74][75][76][77][78][79][80][81].…”

“…The mass spectrum indicates the QA-␤-CD used in these experiments is primarily a mixture of mono-, di-, tri-, and tetra-(2-hydroxyl-3-trimethylammonium)-propyl substituted ␤-CD. Based on ESI-MS results for QA-␤-CD reported in the literature [36,37], albeit from another manufacturer, it is likely that more highly substituted derivatives of QA-␤-CD are present, but below detectable levels. The molecular ion for both I and II is observed at m/z 587 [I or II + Na + ] + (Fig.…”

“…Most of the derivatized CDs are present as mixtures of many isomeric forms differing in the degree of substitution [80,133] and in the position of the substituents [134]. This might become a problem in the sense that any batch-to-batch variability in selector production might limit the reproducibility of the analytical results obtained by them [46,135].…”
