Analysis of IgG Subclass and IgE Response in Allergic Disease Caused by Aspergillus fumigatus by Immunoblotting Techniques

Abstract: Analysis of sera from patients suffering from allergic asthma, extrinsic allergic alveolitis (EAA) due to Aspergillus fumigatus or allergic bronchopulmonary aspergillosis (ABPA) by means of IEF immunoprint and Western blot showed charcteristic IgG subclasses and IgE responses. Asthmatic patients revealed specific IgE antibodies. IgG subclass reactivities in asthmatics and healthy blood donors were weak and uncharacteristic. Major allergens recognized by asthmatic patients have isoelectric points (pI) of pI 4.4… Show more

“…According to Trompelt et al . major allergens of A. fumigatus recognized by IgE antibodies in sera from seven asthmatic patients have molecular weights of 20, 29, 34, 38 and 45 kDa [29]. Proteins recognized with IgE in sera from six patients of ABPA have molecular weights of 14–45 kDa [29].…”

“…The heightened total and specific anti-Aspergillus IgE antibody responses have been described by several groups [14,18,[143][144][145][146][147]. In ABPA, there are also increased amounts of IgG and IgA anti-Aspergillus antibodies, which reflect the Th2 humoral versus Th1 cellular response to Aspergillus antigens in these patients [148][149][150][151][152][153][154][155]. Although other Aspergillus-exposed groups develop IgE, IgG, and IgA anti-Aspergillus antibodies, there is a quantitative increase in IgE anti-Aspergillus antibodies in patients with ABPA.…”

“…Several fungi, including Aspergillus species, have been implicated in allergic asthma. The symptoms due to mold asthma can be distinguished from those attributable to other allergen-induced asthma by history of exposure, skin test reactivity or in vitro demonstration of Aspergillus -specific IgE in patients’ sera [29, 30]. The lack of reliable fungal extracts poses a problem in the development of standardized in vivo or in vitro tests to diagnose A. fumigatus allergy which needs to be discriminated from ABPA for optimal patient management [31, 32].…”
