Analysis of Stained Objects in Histological Sections by Spectral Imaging and Differential Absorption

Abstract: SUMMARYWe describe a new light microscopic imaging system and method to perform high through put color image analysis on histological tissue sections. The system features a computer-controlled, random-access liquid crystal tunable filter and high-resolution digital camera on a conventional brightfield microscope. For any combination of stains, the method determines the spectral transmittance of each stain on the slide and selects two or more wavelengths at which the differential absorption between stain and co… Show more

“…15 A popular system is comprised of coupling a liquid crystal tunable optical filter (LCTF) with a regular light microscope. 16 The LCTF uses electrically controlled liquid crystal elements that transmit certain wavelengths while blocking others and it can operate in the visible range (420 -720 nanometers [nm]) at intervals as narrow as 1 nm. This technology can be used to construct full spectral traces for each pixel in a captured image.…”

“…These colour-based images are commonly captured and displayed as a set of three black and white images collected with red (R), green (G), and blue (B) light, i.e., at wavelengths of approximately 630 nm, 545 nm, and 435 nm, respectively (Ballard & Brown, 1982). These wavelengths are chosen to match the spectral response of the human eye (Irigoyen & Herraez, 2003;Ornberg, Woerner, & Edwards, 1999). The relationship between the hyperspectral tongue images and the colourbased RGB images can be presented by figure 4.…”

“…Johansson et al (33) looked at leukocytes infiltrating rat brain tumor using immunostaining and computerized image analysis. Using spectral ratio of stained and unstained COBs pixels Ornerg (24,28), quantitation of object areas is obtained. Elie et al (40) counted immunostained nuclei using cyclin A by detecting positive pixels and ratio of stained area and total pixel of epithelia of human ovarian adenocarcinomas.…”
