Androgen receptor expression on circulating tumor cells in metastatic breast cancer

Abstract: PurposeAndrogen receptor (AR) is frequently detected in breast cancers, and AR-targeted therapies are showing activity in AR-positive (AR+) breast cancer. However, the role of AR in breast cancers is still not fully elucidated and the biology of AR in breast cancer remains incompletely understood. Circulating tumor cells (CTCs) can serve as prognostic and diagnostic tools, prompting us to measure AR protein expression and conduct genomic analyses on CTCs in patients with metastatic breast cancer.MethodsBlood s… Show more

“…There were both losses (11 of 58 patients (19%)) and gains (13 of 17 patients (76%)) of AR over time. Fujii et al . reported on a small series of 7 mBC patients with AR+ CTCs.…”

“…Here, 60 patients (80%) had AR positive CTCs at baseline. Fujii et al 29 collected whole blood from mBC patients with mostly hormone receptor (HR)+/HER2-primary tumors. They used the Epic Sciences CTC platform to select patient samples with ≥1 CTC on the cytospin.…”

“…However, CTCs are very rare and make up a small minority of cells circulating in blood, so their molecular analysis beyond enumeration is technically very challenging . Various methods to overcome this issue have been under development and evaluation …”

“…6,12 Various methods to overcome this issue have been under development and evaluation. [13][14][15][16][17] The potential of single cell sorting by FACS and whole-genome amplification of CTCs after CellSearch was described previously. 18,19 The main limitation of both of these approaches is that only epithelial cell adhesion molecule (EpCAM)-positive epithelial cells can be isolated and analyzed, because this is the isolation system used by CellSearch.…”
