Aneuploidy, oncogene amplification and epithelial to mesenchymal transition define spontaneous transformation of murine epithelial cells

Abstract: Human epithelial cancers are defined by a recurrent distribution of specific chromosomal aneuploidies, a trait less typical for murine cancer models induced by an oncogenic stimulus. After prolonged culture, mouse epithelial cells spontaneously immortalize, transform and become tumorigenic. We assessed genome and transcriptome alterations in cultures derived from bladder and kidney utilizing spectral karyotyping, array CGH, FISH and gene expression profiling. The results show widespread aneuploidy, yet a recur… Show more

“…Global analysis of gene expression and copy number alterations showed a statistically significant correlation between the average log 2 gene expression and the average log 2 aCGH data in the transformed mouse colon cell lines. Loss of Cdkn2a/p16 is consistent with previous results (Padilla-Nash et al, 2013). 3C).…”

“…In addition, NCI-HPN-F1K Colon showed a statistically significant correlation (R 2 5 0.9657) for a focal deletion of the tumor suppressor gene Cdkn2a/p16 (Supporting Information Figure S2C-E). Loss of Cdkn2a/p16 is consistent with previous results (Padilla-Nash et al, 2013).…”

“…AR amplification has been observed in the Myc-CaP cell line (18). Surprisingly, a number of the chromosomal changes observed in these murine prostate cancer cell lines such as loss of chromosomes 4,9 and gain of chromosomes 11 and 15 are found in immortalized murine cell lines derived from normal kidney or bladder tissue (49). Clearly the selective pressures are different for cells growing in laboratory culture than for cells dividing in the context of a tissue with physiologic tumor microenvironment.…”
