Angiotensin Converting Enzyme and Substance P Changes in Blister Fluid Following Afferent Nerve Stimulation in the Rat

Abstract: We have examined the effect of electrical nerve stimulation on substance P and angiotensin converting enzyme activity in the interstitial fluid of rat skin using a blister model. Following sciatic nerve stimulation, blister fluid immunoreactive substance P (fmol/ml) was increased from 118 (unstimulated side, s.e.m. = 13, n = 15) to 197 (stimulated side, s.e.m. = 26, n = 15, P less than 0.0125, paired t-test, 14 d.f.). Angiotensin converting enzyme (ACE) activity (nmol HL/ml per h) was reduced in blister fluid … Show more

“…Treatment with ACE inhibitors has been reported to restore these blunted responses. 31 -" The mechanism of this effect has not been established but may involve bradykinin. Bradykinin can stimulate the release of both vasodilating prostaglandins as well as endothelium-derived relaxing factor or nitric oxide from endothelial cells.…”

“…30 Inflammatory blister fluid also has a higher content of bradykinin and substance P in animals given ACE inhibitors. 31 There is a strong feeling but little direct evidence that some of the adverse effects of ACE inhibitors such as cough and angioneurotic edema may involve bradykinin. Whether bradykinin, a potent vasodilator, contributes to the acute or chronic hypotensive effect of ACE inhibitors is still controversial.…”

“…The regulation of soluble ACE during inflammation is, thus, not different in these rats. A decrease in soluble ACE also occurs in the blister fluid in the rat blister model (59) or in the plasma of arthritis patients (34).…”
