Biological methods have been used to synthesize silver nanoparticles through materials such as bacteria, fungi, plants, and propolis due to their reducing properties, stabilizer role and environmentally friendly characteristic. Considering the antimicrobial activity of propolis as well as the broad‐spectrum antibacterial effects of silver nanoparticles, this study aim to describe the use of Brazilian propolis to synthesize silver nanoparticles (AgNP‐P) and investigate its antimicrobial activity. The synthesis was optimized by factorial design, choosing the best conditions for smaller size particles. AgNP‐P demonstrated a maximum absorbance at 412 nm in ultraviolet‐visible spectra, which indicated a spherical format and its formation. Dynamic light scattering demonstrated a hydrodynamic size of 109 nm and polydispersity index less than 0.3, showing a good size distribution and stability. After its purification via centrifugation, microscopy analysis corroborates the format and showed the presence of propolis around silver nanoparticle. X‐ray diffraction peaks were attributed to the main planes of the metallic silver crystalline structure; meanwhile infrared spectroscopy demonstrated the main groups responsible for silver reduction, represented by ∼22% of AgNP‐P indicates by thermal analysis. Our product revealed an important antimicrobial activity indicating a synergism between propolis and silver nanoparticles as expected and promising to be an effective antimicrobial product to be used in infections.

BackgroundPropolis is one of the hive products made by bees from wax and plant resins which makes it rich in phytochemicals such as flavonoids and phenolic. The chemical composition of propolis from stingless bees is affected by several factors including vegetation type and species. In Kenya we have diverse landscape vegetation and over 12 stingless bee species. Lifestyle diseases are affecting more people due to unhealthy diets and exposure to harmful chemicals which lead to oxidation in cells. This lead to the production of super radicals which cannot be neutralized only by the endogenous antioxidants in the body. This has led to increased demand for exogenous antioxidants obtained from the diet. Propolis is a known source of natural antioxidants too due to the presence of polyphenols.ResultsIn our study, we determined the total content of flavonoids, phenolic and the antioxidant activity of propolis from six stingless bee species from Kenya. The total flavonoid content (TFC) in all species ranged from 651.90 mg QE/100g and 3262.26mg QE/100g, while the total phenolic content (TPC) ranged from 586.36 mg GAE/100g and 2010.53 mg GAE/100g. Among the species, M. beccarii had the highest concentration of total flavonoids, M. togoensis had high concentrations of total phenolic while M. ferruginea had the highest antioxidant activity.ConclusionThe outcome of this study demonstrates that propolis from Kenyan stingless bees have varying amounts of phytochemicals which is dependent on the species identity. There has been a recent interest in the biosynthesis of nanoparticles using animal systems. Several parts of animals and/or their extracts can serve as good reducing agents for the production of nanoparticle. Animal derivatives like wings of insects, honey and propolis from honeybees, spider silk, nests of paper wasp, wasp venom, silk fibroin, and silk sericin from silkworm have increasingly been explored in the last decade. Different components of chicken eggs like yolk, egg white, and egg shells have also shown promising results. Recently a number of studies have been conducted that utilized different species of earthworms in nanofabrication process. This area requires a lot of consistent efforts since most of the members of animal kingdom have not been explored till now. Urgent efforts are needed to develop this area and enrich the scant knowledge available till date for better utilization in green nanobiotechnology.
