Antibody–Oligonucleotide Conjugates as Therapeutic, Imaging, and Detection Agents

Abstract: Antibody−oligonucleotide conjugates (AOCs) are a novel class of synthetic chimeric biomolecules that has been continually gaining traction in different fields of modern biotechnology. This is mainly due to the unique combination of the properties of their two constituents, exceptional targeting abilities and antibody biodistribution profiles, in addition to an extensive scope of oligonucleotide functional and structural roles. Combining these two classes of biomolecules in one chimeric construct has therefore … Show more

“…Antibodies are considered one of the most effective biomolecules to be used for detection methodologies and applications [42][43][44]. Due to their particular characteristics, such as high affinity and specificity, a large variety of immunoassay formats implementing antibodies as reagents have been developed for disease diagnoses.…”

“…However, similar molecular calculus may be applied to extensively cleared molecular entities which may otherwise have trouble reaching the tissues of interest. A rapidly-advancing molecular space which is typically impeded by delivery issues is that of synthetic therapeutic oligonucleotides including antisense oligonucleotides (ASOs) and short interfering RNA (siRNA) [107][108][109].…”

“…While this Fab'-based study utilized structural cystines for conjugation, other therapeutic antibody-oligonucleotide conjugation systems not yet described in this section have been reported, including two oncology examples which applied two-step conjugations for azide-labeling the antibody which were then treated with cyclooctyne-appended ASOs to generate the conjugates [121,122]. Other antibody-oligonucleotide conjugation methods beyond the scope of this review (i.e., for immuno PCR applications, pre-targeted radiotherapy, or those which utilize non-covalent heterogeneous complexes) have been recently reviewed [109].…”

“…Another major area of application is different detection approaches encompassing mainly immuno-polymerase chain reaction (PCR), proximity ligation/extension assays, electrochemical proximity assay, and DNA-PAINT imaging first and foremost by their exceptional sensitivity. The higher number of potential applications of antibody-oligonucleotide conjugates and antibody-oligonucleotide conjugation techniques have been recently reviewed by Dovgan et al 2 …”
