Anticancer Activity and Mechanism Investigation of Beauvericin Isolated from Secondary Metabolites of the Mangrove Endophytic Fungi

Abstract: One known cyclic peptide, beauvericin, was isolated from the secondary metabolites of mangrove endophytic fungi Fusarium sp. (No. DZ27) in South China Sea. Its structure was determined by spectral analyses and comparisons with reference data from literatures. Beauvericin inhibited growth of KB and KBv200 cells potently with IC50 values of 5.76 ± 0.55 and 5.34 ± 0.09 μM, respectively. Furthermore, beauvericin induced apoptosis through mitochondrial pathway, including decrease of relative oxygen species generati… Show more

“…After that, blots were incubated with horseradish peroxidase (HRP)conjugated secondary antibodies and were visualized with enhanced chemiluminescence as previously described. 36 The antibodies used in this study were as follows: rabbit anti-integrin alpha 2 (ab133557, Abcam), rabbit anti-integrin beta 1 (#9699, Cell Signaling Technology), mouse anti-GAPDH (MB001, Bioworld Technology), rabbit anti-HSP70 (BS6446, Bioworld Technology), rabbit anti-CD9 (#13174, Cell Signaling Technology), mouse anti-CD63 (ab193349, Abcam), rabbit ant-Tsg 101 mAb (E303, Bioworld Technology), HRP-conjugated goat anti-rabbit antibody (#7074, Cell Signaling Technology), and HRP-conjugated goat anti-mouse antibody (BS12478, Bioworld Technology).…”

“…Apoptosis is the process of programmed cell death regulated by a complex network of proliferation and survival genes. Its disequilibrium, i.e., either through the acquisition of anti-apoptotic signals or lack of pro-apoptotic signals, can result in the failure of treatment or a variety of pathological conditions such as cancer or autoimmune and degenerative diseases (18). Therefore, chemical agents that act on molecular targets in apoptotic pathways are likely to be a promising approach for cancer therapy.…”

“…Membranes were blocked with 5% non-fat milk, incubated with primary antibodies at 4°C overnight. After incubation with horseradish peroxidase (HRP) conjugated secondary antibodies for 2 h, blots were revealed by enhanced chemiluminescence procedures according to the manufacturer's protocol [38]. …”
