Anticancer Metal Complexes: Synthesis and Cytotoxicity Evaluation by the MTT Assay

Abstract: Titanium (IV) and vanadium (V) complexes are highly potent anticancer agents. A challenge in their synthesis refers to their hydrolytic instability; therefore their preparation should be conducted under an inert atmosphere. Evaluation of the anticancer activity of these complexes can be achieved by the MTT assay. The MTT assay is a colorimetric viability assay based on enzymatic reduction of the MTT molecule to formazan when it is exposed to viable cells. The outcome of the reduction is a color change of the MT… Show more

“…In addition, similar experiments were performed with human ovarian A2780 cells (parent cisplatin-sensitive cells) and A2780cis cells (with acquired cisplatin resistance), in order to gain more insight into the cytotoxicity pattern of these compounds. The measurements were performed as described previously, [41] by treating the cells with 10 different concentrations of the examined compound dissolved in THF and evaluating the viability of the cells after three days of incubation by use of the methylthiazolyldiphenyltetrazolium bromide (MTT) assay. Compound 1 was administered as a stereoisomeric mixture of the cis and trans isomers.…”

“…All salophan-Ti IV complexes were found to be highly stable, with t 1/2 above 100 h as expected for O-bridged clusters with no labile ligands [15,16,22,23,26]. The cytotoxic activity of the complexes was studied on colon HT-29 cancer cell lines, employing the methylthiazolyldiphenyltetrazolium bromide (MTT) assay [30]. Relative IC 50 values are summarized in Table 2, and cytotoxicity plots are given in Fig.…”
