Antigen presentation and regulatory functions of human monocytes in the in vitro response of lymphocytes against purified protein derivative of tuberculin (PPD)

Abstract: Monocyte functions in the primary and secondary proliferative responses of human lymphocytes against purified protein derivative of tuberculin (PPD) have been studied. By applying the antigens either directly to the responding cells or on antigen-treated autologous monocytes, it was possible to distinguish between two different monocyte functions: antigen presentation and regulation. Their helper function in lymphocyte proliferation was found in cell-free supernatants of autologous and allogeneic monocytes and… Show more

“…The activity of this standard IL 1 was similar in all the tests and, therefore, we chose to express the results as the actual cpm values obtained by the tested supernatants and not by units of activity. (b) Proliferative responses of human lymphocytes: PBM, or MO-depleted PBM were tested for their response against purified protein derivative of tuberculin (PPD) as described [13]. All blood donors for these experiments were positive by a standard Mantoux skin test.…”

“…In some experiments, supernatants were tested for their direct mitogenic activity by using the same culture conditions, but omitting the antigen or mitogen. MO depletion was performed by consecutive removal of plastic-and nylon wool-adherent as well as iron carbonyl phagocytic cells as described [13]. Such depleted cell preparations contained &l% of MO, as measured by phagocytosis of cell latex particles [13].…”

“…Although cells of the monocyte/macrophage lineage are the most significant of the APC (Rosenthal & Shevach, 1973;Treves et al, 1981), other cells types, e.g. dendritic cells (Nussenzweig et al, 1980; Kaye, Chain & Feldman, 1985) and B cells (Gerrard & Fauci, 1982;Issekutz, Chu & Geha, 1982), have been described to perform the same function.…”
