Antigenic determinants of high mobility group chromosomal proteins 1 and 2

Abstract: The antigenic determinants of nonhistone high mobility group chromosomal proteins 1 (HMG-1) and 2 (HMG-2) were studied with rabbit antisera elicited against HMG-1 and against HMG-2 and monoclonal antibodies elicited by HMG-1. The monoclonal antibodies did not distinguish between the two proteins, suggesting that they have specificity toward a shared determinant. Whereas anti-HMG-1 did not, anti-HMG-2 did distinguish between the proteins, suggesting that the anti-HMG-2 serum contains antibodies against peptides… Show more

“…Interestingly, a perinuclear staining pattern of HMG1/HMG2 autoantibodies that develop in patients with refractory ulcerative colitis has been previously reported (50). Autoantibodies to HMG1 and HMG2 have also been found in patients with systemic lupus erythematosis and juvenile rheumatoid arthritis (15,63). One theory is that autoantibodies arise to proteins and other compounds modified during apoptosis.…”

“…In the present study we show that oocytes of various amphibian species contain large amounts of a soluble form of one HMG protein, HMG-A, which is accumulated in the nucleus but is also found in the cytoplasm. We further show that antibodies against mammalian HMG-1 and -2 (32)(33)(34), when microinjected into these nuclei, interfere with transcriptional events in the lampbrush chromosomes.…”

“…antibodies to HMG proteins from calf thymus have been described (19,(32)(33)(34). Immunofluorescence microscopy using cryostat sections through frozen ovaries of adult females of the species studied was essentially as described (28).…”

“…Dating to the seminal studies of Bustin et al in 1982 [122], HMG antibodies have been reported in SLE [122124], mixed connective tissue disease [122], juvenile idiopathic arthritis (JIA) [125–128], canine lupus [129], DIL [130], systemic sclerosis [131], other systemic autoimmune rheumatic diseases [132], primary pulmonary hypertension [133], inflammatory bowel disease [134136], primary biliary cirrhosis [137], type I diabetes [138], autoimmune hepatitis [139], septic shock [140], and liver transplant patients (see Table 2 for more details) [141, 142]. …”
