Malaria is a parasitic infectious disease that remains one of the focuses of world health problems. The ethanol extract of Hedyotis corymbosa has biochemical compounds potentially become a new antimalarial drug. This study aimed to analyze the effect of this extract on parasitemia and hepatomegaly in mice infected with Plasmodium berghei ANKA. Twentyfive BALB/c mice were infected with P. berghei ANKA and grouped into 5 groups. Group 1-3 were treated with 250 mg/Kg BW (HC250), 300 mg/Kg BW (HC300), and 350 mg/Kg BW (HC350) of ethanol extract of H. corymbosa (EEHC), respectively. Group 4 was a positive control (POS) which was given dihydroartemisisnin-piperaquin (DHP) and Group 5 was a negative control (NEG) which was only given CMC Na 1%. Treatments were given orally once a day for four consecutive days. Parasitemia was observed daily on Giemsa-stained tail blood smear. On day 5 the mice were sacrificed, blood were collected by cardiac punctured, the livers were removed and the length, width, and weight were measured. There was no significant difference on parasitemia between Group 1, 2, 3 and NEG. However, the highest inhibition of parasite s growth was found in Group 3 (61.4%. Observation on hepatomegaly, showed that a significant difference on the length of the liver was found between Group 3 and NEG.

Background: All currently available antimalarial drugs are developed from natural product lineages that may be traced back to herbal medicines including quinine, lapachol, and artemisinin. Natural products, which primarily target free radicals or reactive oxygen species, play an important role in the treatment of malaria. Objective: To review role of antioxidative therapy in treatment of malaria by scavenging or counter free radical and also review importance of natural plant extracts as antioxidants in oxidative therapy of malaria treatment. Methods: The search for natural antioxidants was conducted using the following databases of Researchgate, science direct, google scholar, Bentham science using following keywords malaria, reactive oxygen species, natural antioxidants and antiplasmodial. Conclusion: This study reviewed various literature sources related to natural products employed in antimalarial therapy directly or indirectly by countering/scavenging reactive oxygen species that were published between 2016 to till date. The literature survey made it possible to summarize the natural products used in the treatment of malaria, with emphasis on botanical extracts, as a single component, as well as in association with other botanical extracts. Natural antioxidants like polyphenols, flavonoids, alkaloids, having a broad range of biological effects against malaria. This review is pivoted around natural antioxidants obtained from the food and medicinal plants and to explore their application in restraining reactive oxygen species (ROS). We anticipate this article will provide information on future research on the role of antioxidant therapy in malaria infection.

Although many drugs have been discovered to treat malaria infection, many of them face resistance from the host’s body with long-term use. Therefore, this study aimed to evaluate the activity of betalains (from Beta vulgaris) and chloroquine (a reference drug) against brain oxidative stress induced by Plasmodium berghei in male mice. Two protocols were applied in this study: the therapeutic and prophylactic protocols. The results of the therapeutic protocol revealed a significant decrease in the level of parasitemia caused by P. berghei. Additionally, the histopathological changes in various brain regions were markedly improved after treatment with betalains. Regarding the prophylactic protocol, betalains were able to protect the brain tissues from oxidative stress, inflammation, and disrupted neurotransmitters expected to occur as a result of infection by P. berghei. This was demonstrated by modulating the activities of brain antioxidants (SOD and GSH), inflammatory cytokines (IL-6, IL-10, IL-12, TNF-α, and INF-γ), and neurotransmitters (serotonin, epinephrine, and norepinephrine). This study has proven that using betalains as a treatment or as a preventive has a vital and effective role in confronting the brain histopathological, oxidative stress, and inflammatory changes induced by P. berghei infection.
