Antimicrobial Activity and Identification of the Biosynthetic Gene Cluster of X-14952B From Streptomyces sp. 135

Abstract: The bacterial genus Streptomyces is an important source of antibiotics, and genome mining is a valuable tool to explore the potential of microbial biosynthesis in members of this genus. This study reports an actinomycete strain 135, which was isolated from Qinghai-Tibet Plateau in China and displayed broad antimicrobial activity. The fermentation broth of strain 135 displayed strong antifungal activity (>70%) against Sclerotinia sclerotiorum, Botrytis cinerea, Valsa mali, Phytophthora capsici, Glomerell… Show more

“…Germicidin A, a pyranone autoregulator of sporulation [ 49 ], was putatively present in the extracts from N. alba (PMIC_1A11B.2 and PMIC_2A11B.1). The antibiotic X-14952B, a 20-membered macrolide lactone with broad-spectrum antibacterial properties [ 50 , 51 ], was found in the extracts of N. alba PMIC_2H2C.2 and PMIC_2A11B.1. Also present in the extract of N. alba PMIC_1F6A.3 was cyclo(Tyr-Leu), a diketopiperazine that exhibits antibacterial [ 52 ] and antifungal [ 53 ] activities.…”

“…Physiological characteristics such as temperature tolerance, NaCl tolerance, and pH level were investigated. Various enzymatic activity tests like catalase, oxidase, nitrate reductase, amylase, caseinase, gelatin hydrolysis citrate utilization, and melanin production tests were also performed according to the previous description in [ 9 , 18 ].…”

“…Currently, the majority of the commercially and medically known antibiotics have originated from a different genus of actinomycetes [ 6 , 7 ]. Actinomycetes have also been the source of many industrially and clinically important bioactive metabolites in treating cancer, inflammatory diseases, allergies, anthelmintics, autoimmune diseases, infections, and others [ 8 , 9 ]. Among actinomycetes genera, Streptomyces are the largest genus and the most frequent producer of antibiotics which is continuously being explored for antimicrobial drug discovery [ 9 , 10 ].…”
