Purpose: This research aimed to investigate the antibacterial activity and potential mechanism of luteolin against T. pyogenes. Materials and Methods: The broth microdilution method was used to determine the minimum inhibitory concentrations (MICs) of luteolin against various T. pyogenes strains. The potential mechanism of action of luteolin was elucidated through testing and analysing the luteolin-induced alterations of T. pyogenes in several aspects, including cell wall, cell membrane, protein expression, nucleic acid content, topoisomerase activity and energy metabolism. Results: The MIC values of luteolin against various T. pyogenes isolates and ATCC19411 were 78 µg/mL. The increased cell membrane permeability, destruction of cell wall integrity and TEM images after exposure to luteolin showed that the cell wall and membrane were damaged. The content of total protein and nucleic acid in T. pyogenes decreased significantly after treatment with luteolin (1/2 MIC) for 12, 24, and 36 h. Moreover, a hypochromic effect was observed in the absorption spectrum of luteolin when deoxyribonucleic acid (DNA) was added. In addition, after treatment with luteolin, a decrease in nicked or relaxed DNA content, which was catalysed by T. pyogenes-isolated DNA topoisomerase, was observed. In addition, the adenosine triphosphate (ATP) content in cells and the activity of succinate dehydrogenase (SDH) both decreased when T. pyogenes was exposed to different concentrations (1/4 MIC, 1/2 MIC, 1 MIC, 2 MIC) of luteolin for 1 h. Conclusion: Luteolin showed distinct antibacterial activity against T. pyogenes by multiple actions, which mainly include destroying the integrity of the cell wall and cell membrane, influencing the expression of proteins, inhibiting nucleic acid synthesis, and interfering with energy metabolism.

Article history Trueperella pyogenes is a pathogenic bacterium that causes suppurative infections in domestic and wild animals as well as humans. This paper describes a clinical case of subcutaneous abscess by T. pyogenes in a bovine in the Western Amazon, Brazil. During anamnesis, intramuscular injectable drug administration in the middle third of the left gluteal biceps muscle and the practice of reusing needles were reported. The clinical examination revealed edema, hyperthermia and high pain sensitivity to the touch in the region. A sample was collected through a puncture incision for the bacterial culture, during which T. pyogenes was isolated. The therapeutic approach was the use of a systemic antimicrobial and topical care, with a good prognosis. This report is the first description of infection by T. pyogenes in cattle in the Western Amazon and underscores the importance of appropriate sanitary management in herds, especially the use of disposable needles and hygiene at the site of parenteral drug application.

True.pe.rel'la. N.L. fem. n. Trueperella named after Hans Georg Trüper, the German microbiologist. Actinobacteria / Actinobacteria / Actinomycetales / Actinomycetaceae / Trueperella Cells are Gram‐reaction‐positive, nonmotile, non‐spore‐forming coccobacilli and short rods that occur singly, in pairs (V, T, and palisade formations), or in clusters. Cells vary in shape and size in different media. Cells from 24‐h‐old broth cultures are Gram‐reaction‐positive but may be Gram‐reaction‐variable in older cultures. Pinpoint, in most cases β‐hemolytic colonies occur on sheep blood agar after 24 h of incubation. After 48–72 h of incubation, colonies are 0.5–1.5 mm in diameter, convex, circular, and translucent with entire edges. Aerobic and facultatively anaerobic. Strictly fermentative. Lactic acid is the primary metabolic product in glucose–yeast extract broth, but acetate and succinate are minor products. The peptidoglycan is of the type A5a, variations l ‐Lys– l ‐Ala– l ‐Lys– d ‐Glu or l ‐Lys– l ‐Lys– d ‐Glu. The muramic acid residue of the peptidoglycan is N ‐acetylated. Characteristic whole‐cell sugars are rhamnose and glucose. Mycolic acids are not present. The long‐chain fatty acids are primarily straight‐chain saturated and monounsaturated in addition to traces of branched‐chain fatty acids of the iso‐/anteiso type. The phospholipid type is type PI, comprising diphosphatidylglycerol (DPG), phosphatidylglycerol (PG), phosphatidylinositol (PI), and an unknown phosphoglycolipid (PGL) of unknown composition. The principal menaquinone is MK‐10(H4). DNA G + C content (mol%) : 56–66. Type species : Trueperella pyogenes Yassin et al. 2011 VP .
