Phaeodactylum tricornutum is a model diatom with numerous potential applications in the industry, including the production of high-value carotenoid pigments such as fucoxanthin. This compound is a potent antioxidant currently extracted mainly from brown macroalgae. Fucoxanthin exhibits several biological properties with well-known beneficial effects in the treatment and prevention of lifestyle-related diseases. P. tricornutum offers a valuable alternative to macroalgae for fucoxanthin production as it has a specific productivity that is 10-fold higher as compared with macroalgae. However, production processes still need to be optimised to become a cost-effective alternative. In this work, we investigated the optimal supplementation of nitrate in a cultivation medium that is currently used for P. tricornutum and how this nitrate concentration affects cell growth and fucoxanthin production. It has previously been shown that the addition of sodium nitrate increases productivity, but optimal conditions were not accurately determined. In this report, we observed that the continuous increase in nitrate concentration did not lead to an increase in biomass and fucoxanthin content, but there was rather a window of optimal values of nitrate that led to maximum growth and pigment production. These results are discussed considering both the scale up for industrial production and the profitability of the process, as well as the implications in the cell’s metabolism and effects in fucoxanthin production.

Küresel ısınmaya bağlı olarak su kaynaklarının azalması ve temiz suya ulaşmada yaşanan zorluklar günümüzde önemli sorunlar olarak insanların karşısına çıkmaktadır. Önemi bu gibi nedenlerle daha da artan su kaynakları ve bu su kaynaklarında yaşayan canlılarla ilgili yapılan yeni çalışmalar, bu kaynakların durumu hakkında bilgiler sağlamaktadır. Algler yaşadıkları ortama oksijen sağlamaları ve besin zincirinin ilk basamağını oluşturmaları nedeniyle ortamda bulunan diğer canlıların yaşamlarını sürdürebilmesi için gerekli organizmalardır. Algler, içinde bulundukları sucul ekosistemlerde birincil üretici olmaları nedeniyle oldukça önemli canlılardır. Bu önemlerinden dolayı mikroalgler üzerinde birçok çalışma yapılmaktadır. Bu çalışmada, Konya şehrinin önemli içme suyu kaynaklarından biri olan Altınapa Baraj Gölü'nün bentik alg florasından izole edilen bazı yeşil alg türlerinin klorofil-a, klorofil-b, toplam karotenoid içeriği ve antioksidan aktivitelerinin belirlenmesi amaçlanmıştır. Kültür koşullarında yüksek çoğalma potansiyeli gösteren üç Chlorophyta (Chlorella vulgaris, Acutodesmus obliquus, Monoraphidium minutum) türü seçilmiş ve çalışmalar bu türlerle gerçekleştirilmiştir. Chlorophyta bölümüne ait Monoraphidium minutum türü referans alınan antioksidana yakın IC50 değerleri verdiğinden, uygun koşullarda üretildiğinde gıda ve eczacılık endüstrilerinde ekonomik katkı sağlayabileceği sonucuna varılmıştır.

Heavy metals is a collective term describing metals and metalloids with a density higher than 5 g/cm3. Some of them are essential micronutrients; others do not play a positive role in living organisms. Increased anthropogenic emissions of heavy metal ions pose a serious threat to water and land ecosystems. The mechanism of heavy metal toxicity predominantly depends on (1) their high affinity to thiol groups, (2) spatial similarity to biochemical functional groups, (3) competition with essential metal cations, (4) and induction of oxidative stress. The antioxidant response is therefore crucial for providing tolerance to heavy metal-induced stress. This review aims to summarize the knowledge of heavy metal toxicity, oxidative stress and antioxidant response in eukaryotic algae. Types of ROS, their formation sites in photosynthetic cells, and the damage they cause to the cellular components are described at the beginning. Furthermore, heavy metals are characterized in more detail, including their chemical properties, roles they play in living cells, sources of contamination, biochemical mechanisms of toxicity, and stress symptoms. The following subchapters contain the description of low-molecular-weight antioxidants and ROS-detoxifying enzymes, their properties, cellular localization, and the occurrence in algae belonging to different clades, as well as the summary of the results of the experiments concerning antioxidant response in heavy metal-treated eukaryotic algae. Other mechanisms providing tolerance to metal ions are briefly outlined at the end.
