Panax bipinnatifidus Seem. (Araliaceae), 1 of 11 kinds of ginseng ( Panax spp.), has been used in traditional medicine but there are very few publications regarding its chemical constituents. This first study on phytochemical components in the aerial parts of the title ginseng led to the isolation of 7 oleanane-type saponins (1-7) including 1 new compound (1), together with hemsloside H1 (2), stipuleanoside R1 (3), stipuleanoside R2 (4), araloside A (5), and oleanolic acid 28- O-β-d-glucopyranosyl ester (6), and 3- O-[β-d-xylopyranosyl(1→2)-β-d-glucuronopyranosyl]oleanolic acid 28- O-β-d-glucopyranosyl ester (7), respectively. The structure of the new saponin was elucidated as 3- O-{β-d-xylopyranosyl(1→3)-[α-l-arabinofuranosyl(1→4)]-β-d-glucopyranosyl}oleanolic acid 28- O-β-d-glucopyranosyl ester (1) on the basis of extensive evidence of chemical and spectroscopic experiments. The obtained results significantly contribute to the phytochemical database of P. bipinnatifidus associated with the chemotaxonomic meaning of P. bipinnatifidus and Panax spp.

Panax bipinnatifidus Seem. belonging to the family Araliaceae has been used as a folk medicine in Viet Nam. Our ongoing study on bioactive compounds from Panax bipinnatifidus Seem. collected in Sapa, Lao Cai resulted in the isolation of two saponins together with the well-known saponin Stipuleanoside R2. Their structures were identified as 3-O-β-D-glucopyranosyl-(1→3)-β-D-glucuronopyranosyloleanolic acid (1) and Stipuleanoside R1 (2) based on NMR spectroscopic data as well as comparison with the literature data. For the first time, 3-O-β-D-glucopyranosyl-(1→3)-β-D-glucuronopyranosyloleanolic acid (1) was found in genus Panax. On biological screening, the crude extract and the principal saponin, stipuleanoside R2 showed weak inhibitory effect on markedly nitric oxide production in lipopolysaccharide-treated RAW 264.7 cells.

Amylase inhibitors represent a drug target for lowering postprandial hyperglycemia in diabetic patients. According to traditional medicines, Panax plants have been utilised to treat diabetes and considered as a valuable source to develop anti-diabetic supplementary products. The current study aimed to evaluate the α-amylase inhibitory activities of Panax bipinnatifidus root cultivated in Vietnam and its main chemical constituents. The two major saponins, Araloside A and Stipuleanoside R2 were confirmed by ultra performance liquid chromatography quadruple-time of flight-mass spectrometry (UPLCQ-TOF/MS) and nuclear magnetic resonance (NMR). Then, the results indicated that Araloside A and Stipuleanoside R2, were shown to moderately inhibit the α-amylase activity in an enzymatic assay with IC50 values of 781.46 and 2746.90 μg/mL, respectively. Moreover, Araloside A (Vmax ~ 0.0635 µM/min, Km ~ 4.691 µM) was displayed as a completive inhibitor of α-amylase in the kinetic study whereas Acarbose (Vmax ~ 0.053 µM/min, Km ~ 6.521 µM) showed a mixed inhibition. This study suggested the posibility of Araloside A acting as an α-amylase inhibitor. Also, it supports the traditional use of P. bipinnatifidus in antidiabetic dietary.
