Apigenin Induced Apoptosis by Downregulating Sulfiredoxin Expression in Cutaneous Squamous Cell Carcinoma

Abstract: Cutaneous squamous cell carcinoma (cSCC) is the second carcinoma in nonmelanoma skin cancer (NMSC). Sulfiredoxin (Srx) is an antioxidant protein with a role in maintaining redox homeostasis. And Srx has an oncogenic role in skin tumorigenesis. In the current study, we found that apigenin, as a natural flavonoid, downregulated the expression of Srx protein in cSCC cell lines. Apigenin also inhibited the ability of cell proliferation and migration and induced apoptosis in cSCC cell lines. Our results also showed… Show more

“…Apigenin has a noteworthy role in apoptosis induction. It increased apoptosis through the activation of the MAPK signaling pathway, as well as through the reduction in sulfiredoxin expression [ 68 ]. Apigenin was demonstrated to increase poly-(ADP-ribose) polymerase (PARP) proteolytic cleavage and to trigger a rapid enhancement of caspase-3 activity.…”

“…Another target of apigenin in SCC is Sulfiredoxin (Srx), a protein with an oncogenic role in skin tumorigenesis. A study by Wang et al showed that apigenin induces apoptosis by downregulating Srx through the MAPK pathway in SCC cells [ 86 ]. Regarding the regulation of oxidative stress, Paredes-Gonzalez et al found that apigenin can reverse the hypermethylation of 15 CpG sites in the Nrf2 promoter in skin epidermal JB6 P+ cells.…”

“…Zhang et al observed that the use of resveratrol as an adjuvant for PDT potentiates the effect of tumor growth inhibition in cells with SCC; the pathway involved would appear to be that of p38/MAPK [ 86 ].…”

“…Apigenin has been shown to have a significant part in causing apoptosis. This is accomplished by triggering the MAPK signaling pathway and reducing sulfiredoxin expression (Wang et al, 2022 ). Studies have demonstrated that apigenin increases the cleavage of poly‐(ADP‐ribose) polymerase (PARP) and rapidly enhances caspase‐3 activity, resulting in DNA fragmentation and apoptosis (Ghanbari‐Movahed et al, 2023 ; Gupta et al, 2002 ).…”
