Apolipoprotein D is Elevated in Oligodendrocytes in the Peri-Infarct Region after Experimental Stroke: Influence of Enriched Environment

Abstract: Injury to the brain (e.g., stroke) results in a disruption of neuronal connectivity and loss of fundamental sensori-motor functions. The subsequent recovery of certain functions involves structural rearrangements in areas adjacent to the infarct. This remodeling of the injured brain requires trafficking of macromolecular components including cholesterol and phospholipids, a transport carried out by apolipoproteins including apolipoprotein D (apoD). We investigated the changes in the levels of apoD mRNA and pro… Show more

“…81,82) Apo D is synthesized in oligodendrocytes and astrocytes of the CNS 83) and shows neurotrophic and synaptogenic effects in dorsal root ganglion neurons in vitro. 84) The levels of apo D are increased in the pre-frontal cortex of AD patients 85) and in brains of an animal model of Niemann-Pick type C disease, 86) without any change in the amount of apo E. The mRNA level and immunoreactivity of apo D is up-regulated in oligodendrocytes and astrocytes in the peri-infarct area of experimental stroke, 87,88) and the level of apo E is also increased in this area of animal stroke model. 89) The level of apo D in the brain of apo E knockout mice was increased 50-fold compared to that in wild type mice.…”

“…In shamoperated animals, equivalent brain regions were dissected, also designated as IC and PI (a schematic representation of IC and PI is included in the Results section). Whole cellular protein extractions from mice brains were performed as in Rickhag et al (2008). Interleukin-1b levels were determined by an electrochemiluminescence-coupled immunosorbent assay according to the manufacturer's instructions and using a SECTOR Imager 6,000 (Meso Scale Discovery, Gaithersburg, MD, USA).…”

“…12 Likewise, enriched housing initiated days after experimental stroke enhances long-term functional recovery, which occurs without decreasing infarct size. Here, the beneficial processes stimulate glia cells to encapsulate the infarct [13][14][15] or enhance neuronal plasticity. 16 Importantly, enriched housing has a profound effect on the inflammatory reponse after brain injury 14 and modulates cell genesis.…”
