Apolipoprotein E influences melatonin biosynthesis by regulating NAT and MAOA expression in C6 cells

Abstract: Previous studies have demonstrated that apolipoprotein E (ApoE) genotype and melatonin are closely associated with Alzheimer's disease (AD). However, the relationship between ApoE genotype and melatonin remains unclear. Recently, we reported that cultured rat cortical astrocytes and glioma C6 cells synthesize melatonin. In the current study, we investigated the effect of ApoE genotype on melatonin biosynthesis. C6 cells with stable expression of ApoE isoforms (ApoE 2, 3 and 4) were established. A higher level … Show more

“…Melatonin (N‐acetyl‐5‐methoxytryptamine) is synthesized by the pineal gland or extra‐pineal tissues that exerts various biological effects, especially anti‐inflammatory and immunomodulatory functions . Melatonin is synthesized from tryptophan (Trp) by a highly conserved pathway involving the conversion of serotonin successively by two key enzymes such as N‐acetyltransferase (NAT) and HIOMT .…”

“…A major breakthrough was the demonstration that the developing brain produces its own melatonin even in pinealectomized rats . Although an increasing number of publications have documented the expression of melatonin‐synthesizing enzymes as well as high levels of melatonin in the brain, melatonin production was only effectively demonstrated in cultured astrocytes and C6 rat glioma cells . Thus, concerns have been raised that question which other nonpineal cells could be responsible for melatonin synthesis.…”

“…Cultured cortical astrocytes from neonatal rats were shown to synthesize melatonin (1) at rates attaining about one third of those of pinealocytes under same conditions [54]. Melatonin (1) formation was also demonstrated in C6 rat glioma cells [54,55]. The role of glial melatonin (1) deserves further attention.…”
