Apoptosis and DNA fragmentation precede TNF‐induced cytolysis in U937 cells

Abstract: The hypothesis that activation of apoptosis and DNA fragmentation is involved in TNF-mediated cytolysis of U937 tumor cells was investigated. Morphological, biochemical, and kinetic criteria established that TNF activates apoptosis as opposed to necrosis. Within 2-3 h of exposure to TNF, U937 underwent the morphological alterations characteristic of apoptosis. This was accompanied by cleavage of DNA into multiples of nucleosome size fragments. Both of these events occurred 1-2 h prior to cell death as defined … Show more

“…Samples were mounted and subjected to microscopic analysis. Nuclear condensation as revealed by DAPI staining in the absence of trypan blue uptake is considered characteristic of apoptosis as opposed to necrosis (14,15). The quantitative analysis included counting the number of apoptotic relative to all identifiable cells from at least 10 microscopic fields with an average of 70 cells per field.…”

“…This finding is consistent with the findings for this cell line reported from our and other laboratories. 9,11 An apoptosis-inducing effect of the optimal concentration of TNF (10 ng/ml) was rapid and potent, and maximal effect was obtained at 2 days of incubation with TNF, when apoptosis by GM-CSF was significant (Figure 1b) but only minimal. Under these conditions, potent apoptosis-inducing effect of TNF was further potentiated by the simultaneous addition of GM-CSF with TNF ( Figure 2).…”

“…The percentage of apoptotic cells was evaluated by a modification of the JAM test (Matzinger, 1991) and of the quantitative assay described by Wright et al (1992). Briefly, NB cells were treated or not with IFN-g, as described, in 24 well tissue culture plates.…”
