Application of a Static Fluorescence‐based Cytometer (the CellScan) in Basic Cytometric Studies, Clinical Pharmacology, Oncology and Clinical Immunology

Abstract: The CellScan apparatus is a laser scanning cytometer enabling repetitive fluorescence intensity (FI) and polarization (FP) measurements in living cells, as a means of monitoring lymphocyte activation. The CellScan may serve as a tool for diagnosis of rheumatoid arthritis (RA) and systemic lupus erythematosus (SLE) as well as other autoimmune diseases by monitoring FP changes in peripheral blood lymphocytes (PBLs) following exposure to autoantigenic stimuli. Changes in FI and FP in atherosclerotic patients' PBL… Show more

“…Suspension cells, such as leukocytes, in particular, are especially difficult to analyze since these cells are constantly mobile in the medium . The microcavity array provides an environment that allows trapped cells to be cultured or maintained by transporting nutrients and oxygen by microfluidic networks, which is an important aspect in cellular activity assays , and can fulfill the requirements needed for precise cell component measurement and long-term monitoring of cells . Finally, unlike systems that have been elucidated using stable cell lines or model cell samples, , we believe that applying real samples is of great importance and provides us with better insights on the applicability and feasibility of our technology.…”

“…An increase of Rh123 dyed cells polarization versus the control cells indicates the induction of early apoptotic process in tumor cells, while decrease of FP indicates the lack of programmed cell death [24,25]. For every palladium compound, a hyperpolarization was observed, the most significant increase being for compound 2, and compounds 1 and 3 exhibit lower apoptotic effects (Table 3).…”

“…The CellScan system is a laser scanning equipment to measure fluorescence intensity (FI) and polarization (FP) of cells labeled with fluorescent dyes. An important constituent of the system is the ''cell carrier", a grid with apertures dimensioned to hold one single cell, which allows the FI and FP measurement of each cell individually [25]. An increase of polarization values in FDA stained cells, indicates the cell proliferation, while hyperpolarization of Rh123 is a marker of early apoptosis.…”

“…The Cellscan is a static laser scanning cytometer system, developed to perform highly precise sequential fluorescence intensity and polarization measurements of probes within intact living cells, under physiological conditions 19 . Since this system was first introduced by Deutsh and Weinreb in 1994 20 it has been widely used to detect activation of lymphocytes both in physiological 21 and pathological situations including cancer, autoimmune disorders and atherosclerosis 22–25 . In 2005, the Cellscan has been introduced as a novel in vitro test to diagnose cutaneous ADR 26,27 .…”
