Application of Experimental Design Methodologies in the Enantioseparation of Pharmaceuticals by Capillary Electrophoresis: A Review

Abstract: Chirality is one of the major issues in pharmaceutical research and industry. Capillary electrophoresis (CE) is an interesting alternative to the more frequently used chromatographic techniques in the enantioseparation of pharmaceuticals, and is used for the determination of enantiomeric ratio, enantiomeric purity, and in pharmacokinetic studies. Traditionally, optimization of CE methods is performed using a univariate one factor at a time (OFAT) approach; however, this strategy does not allow for the evaluati… Show more

“…This separation technique presents a series of advantages such as a fast method development, short analysis time, low consumption of analytes and reagents and, when compared to LC, a lower use of organic solvents, being considered a "greener" technique. Moreover, for chiral separation, CE provides a high separation efficiency, a wide variety and flexibility of chiral selectors as well as a lower consumption of them [138].…”

“…Capillary electrophoresis (CE) counts as a complementary technique to HPLC and represents a powerful alternative for separation of enantiomers by means of simple method development and low sample and electrolyte consumption combined with inexpensive UV detection. In this field, previous work has already shown the potential of cyclodextrins as chiral additives in CE [19][20][21]. This chiral selector class comprises 6 (α-), 7 (β-) or 8 (γ-cyclodextrin) cyclic glycopyranose units forming a truncated cone.…”

“…Sensitivity analysis was performed based on the aforementioned 27 experiments to determine the design variables that affect the output variables [24]. Figure 5 shows that the torque ripple of the objective function is the most affected by the barrier length (X3).…”
