Aptamer-conjugated graphene oxide-based surface assisted laser desorption ionization mass spectrometry for selective extraction and detection of Aβ1–42 in an Alzheimer’s disease SH-SY5 cell model

Abstract: The generation and accumulation of amyloid-beta peptide (Aβ1–42) in amyloid plaques are key characteristics of Alzheimer’s disease (AD); thus, specific detection of Aβ1–42 is essential for the diagnosis and treatment of AD. Herein, an aptamer-conjugated graphene oxide (Apt-GO) sensor was synthesized by π-π and hydrophobic interactions using thiol poly (ethylene glycol) amine (SH-PEG-NH2) as a spacer unit. Then, it was applied to selective capture of Aβ1–42, and the resulting complex was directly analyzed by su… Show more

“…In addition to Tau, Takahashi, and colleagues [ 85 ] and Song group [ 86 ] successfully selected RNA aptamers against the Aβ1-40 peptide, showing that the aptamers efficiently inhibit Aβ aggregation in vitro with therapeutic potential for treating AD. More recently, Lida et al focused on preventing AD by destroying the formation of prion protein-Aβ oligomers [ 102 ].…”

“…Bonanni et al (2012) first examined the ability of GO to conjugate with DNA hybrids and discovered its promising role in DNA labeling. Song et al (2022) conjugated GO with Aβ 1-42 -specific aptamer to generate a novel biosensor that could detect Aβ 1-42 of 0.1 to 10 uM through surface-assisted laser desorption ionization mass spectrometry. MiRNA-137 was identified as a reliable biomarker for AD (Azimzadeh et al, 2017).…”
