Arabidopsis myrosinases link the glucosinolate-myrosinase system and the cuticle

Abstract: Both physical barriers and reactive phytochemicals represent two important components of a plant’s defence system against environmental stress. However, these two defence systems have generally been studied independently. Here, we have taken an exclusive opportunity to investigate the connection between a chemical-based plant defence system, represented by the glucosinolate-myrosinase system, and a physical barrier, represented by the cuticle, using Arabidopsis myrosinase (thioglucosidase; TGG) mutants. The tg… Show more

“…Considering the presence of GLSs in guard cells (Geng et al., 2016; Zhu and Assmann, 2017), how the GM system plays a role in guard cell functions (e.g., stomatal immunity) is an interesting question. Clearly, mutation of the TGG1 and/or TGG2 genes affected the guard cell size, stomatal aperture, and leaf metabolites, such as fatty acids, glucosinolates, and indole compounds (Ahuja et al., 2016). Another proteomic study of trichome and epidermal pavement cells did not identify the TGG1 protein in the samples (Wienkoop et al., 2004).…”

“…The glucosinolates are a group of secondary metabolites involved in ABA-regulated stomatal opening [ 104 ] and floral development in drought conditions [ 105 ]. The tgg1 tgg2 mutant showed stomata with closed aperture in leaves resembling plants in the face of drought stress [ 106 ]. Thus, carbohydrate hydrolysis can also be involved in ICE1-regulated anther dehydration.…”

“…For SEM analysis, tissues were dissected under anatomical lens (SMZ-161-BLED, Motic, China) if needed, then immediately mounted on aluminum stubs for SEM. For leaf tissues, small pieces (d = 8 mm) of leaves from about 5-week-old plants were cut, fixed, dehydrated and coated as previously described [ 106 ]. These images were taken with scanning electron microscope TM3000 (TM3000 Tabletop Microscope, HITACHI, Japan).…”

“…Myrosinases are involved in the plant defense system and work in conjunction with glucosinolates produced by Brassicaceae species against biotic diseases. The dual glucosinolate-myrosinase system acts against biotic defense by catalyzing the breakdown of the glucosinolates upon plant damage, thereby producing hydrolysis products, which are toxic to pathogens and herbivores, and acting as a plant defense system to certain microbial pathogens [ 36 ].…”
