Arachidic Acid in Extender Improves Post‐thaw Parameters of Cryopreserved Nili‐Ravi Buffalo Bull Semen

Abstract: Cryopreservation process reduces lipids and phospholipids from buffalo bull spermatozoa. It was therefore hypothesized that supplementation of fatty acid to extender may improve the post-thaw quality of buffalo semen. The objective was to evaluate the effect of arachidic acid supplementation in extender on post-thaw quality of buffalo bull (Bubalus bubalis) spermatozoa. Semen was collected from three adult Nili-Ravi buffalo bulls of similar age group with artificial vagina (42°C) for 3 weeks (replicate). Quali… Show more

“…More recently, addition of 8% coconut oil as a source of lauric acid to egg yolk based-extender (79), 5 ng/ml α-linoleic acid to the BioXcell ® (80), and 20 ng/ml arachidonic acid to tris-citric acid extender (81) enhanced quality of sperm following cryopreservation. Additionally, Iodixanol is commonly used as a medium for density gradient centrifugation.…”

“…Oxidants, such as reactive oxygen species (ROS) which are produced physiologically in living cells during respiration as well as by abnormal or dead sperm and phagocytic cells of both the ejaculate and female reproductive tract, affect the quality of postthawed sperm in animals [7,8]. These ROS can inhibit sperm motility, capacitation, and acrosome reaction mediated by lipid peroxidation (LPO) of sperm membrane.…”

“…Sperm chromatin integrity was assessed as practiced by Ejaz et al (2014). Air dried smears of semen samples were fixed in 96% ethanol-acetone (1:1) at 4°C for 30min, hydrolyzed with 4N HCl at 25°C for 10-30 min.…”
