Areca nut components stimulate ADAM17, IL-1α, PGE2 and 8-isoprostane production in oral keratinocyte: role of reactive oxygen species, EGF and JAK signaling

Abstract: Betel quid (BQ) chewing is an etiologic factor of oral submucous fibrosis (OSF) and oral cancer. There are 600 million BQ chewers worldwide. The mechanisms for the toxic and inflammatory responses of BQ are unclear. In this study, both areca nut (AN) extract (ANE) and arecoline stimulated epidermal growth factor (EGF) and interleukin-1α (IL-1α) production of gingival keratinocytes (GKs), whereas only ANE can stimulate a disintegrin and metalloproteinase 17 (ADAM17), prostaglandin E2 (PGE2) and 8-isoprostane pr… Show more

“…Extensive epidemiologic evidence have demonstrated increased risk for the development of OSCC associated with areca nut chewing in Southeast Asia [2, 3]. Mounting short-term assay studies have demonstrated that arecoline, a major areca nut alkaloid, contributes pathogenesis of OSCC [4, 5].…”

“…Higher level of 8-OHdG has been noted in smokers than non-smokers 30 . It has been demonstrated that reactive oxygen species mediated DNA double strand breaks and 8-OHdG occurs via secretory cytokines in areca-nut exposed oral keratinocytes 41 . With this rational in view, validation of the present in silico observations was sought to be done by quantifying the level of 8-OHdG in DNA digests of cancer cells obtained from patients with different GSTP1 genotypes.…”

“…Inducing cytotoxicity, genotoxicity and inflammatory mediators’ release by a number of chemicals such as cadmium, areca nut, resin monomers and camphorquinone are associated with cellular ROS production [12,17,20,35]. The sources of ROS can be derived from host cells and microorganisms such as Porphyromonas gingivalis and Enterococci faecalis [3638].…”
