Arterial chemoembolization with cisplatin microcapsules

Abstract: Summary Cisplatin (CDDP) was microcapsulated with ethylcellulose. Sustained release of CDDP from the microcapsule, particularly non-protein-bound CDDP, which should have antitumour activity, was demonstrated by an in vitro test. Using a bioassay, it was proven that the biological activity of CDDP was not affected by the microencapsulation process. When CDDP-mc were infused into the maxillary artery of patients with carcinoma of the maxillary sinus or oral cavity, the CDDP level in the circulating blood was sign… Show more

“…The total leukocyte count of cisplatin as well as niosomal cisplatin treated group on day 15 showed significant reduction in number of leukocytes by 34.12% and 15.72 % respectively (Table a b c 3) when compared to their initial blood cell count of day 0 (the day 0 leukocyte count acts here as a self control). As far as the weight of the animals is concerned, in control group, 13.04% weight gain was observed on day 15, and 6.075% weight gain was observed in empty niosomes treated group as compared to their initial weights ( Table 4).…”

“…These are the reasons why chemoembolization was used rarely in the past. According to the body of literature, only 54 head and neck SCC patients were treated using embolization protocols in Japan and China in a period of more than 14 years (Okamoto et al, 1985(Okamoto et al, , 1986Kato et al, 1996;Tomura et al, 1996;Konno et al, 1998;Tomura et al, 1998;Li et al, 1999).…”

“…Firstly, cytotoxic drugs have been co-administered with biodegradable microspheres which temporarily slow hepatic arterial blood-flow in the tumourbearing liver and increase uptake of drug by the cells (Dakhil et al, 1982;Thom et al, 1989). Secondly, anti-cancer agents, including Adriamycin, mitomycin C, 5 fluorouracil, cis-platin and cytocidal radio-nuclides have been loaded into particles which act as controlled release vehicles in the target tissue Fujimoto et al, 1985;Okamoto et al, 1986;Herba et al, 1988).…”
