Aryl Hydrocarbon Receptor: A New Player of Pathogenesis and Therapy in Cardiovascular Diseases

Abstract: The aryl hydrocarbon receptor (AhR) is a DNA binding protein that acts as a nuclear receptor mediating xenobiotic metabolism and environmental responses. Owing to the evolutionary conservation of this gene and its widespread expression in the immune and circulatory systems, AhR has for many years been almost exclusively studied by the pharmacological/toxicological field for its role in contaminant toxicity. More recently, the functions of AhR in environmental adaption have been examined in the context of the o… Show more

“…There is an extensive crosstalk between AhR and the proinflammatory transcription factor NF-κB [108], and AhR may also be directly involved in the transcriptional regulation of proinflammatory genes [109,110,111,112,113,114]. AhR and PAHs also appear to play a central role in CVD, as reviewed elsewhere [115,116,117], providing a potential link between PM exposure and adverse effects. These are other possible components and effects not picked-up by acellular assays for OP.…”

“…Thus, inhibition of the NF-kB signaling pathways may reduce inflammation, attenuate atherosclerosis and prevent its complications (Pamukcu et al, 2011). There is also evidence that AhR plays a role in the pathogenesis of cardiovascular diseases (Yi et al, 2018). This may be due to its ability to modulate biological responses or other signaling pathways within different cells types, e.g., macrophages, in the cardiovascular system (Hewitson et al, 2004;Ogando et al, 2016;Wang et al, 2017).…”

“…Cytochromes P450 and in particular CYP1A1 are of crucial importance for the metabolic activation and detoxification of many xenobiotics, such as PAHs, including through the ARNT pathway (AHR/aryl hydrocarbon nuclear translocator). Modulating the expression of this bioactivation enzyme may be an appropriate strategy to prevent toxic effects and the development of CVDs, inflammation, and cancer [87,88]. In this context, we investigated the influence of microalgal extracts on the metabolism of B[a]P and, in particular, the expression of CYP1A1.…”
