Aryl Polyenes, a Highly Abundant Class of Bacterial Natural Products, Are Functionally Related to Antioxidative Carotenoids

Abstract: Bacterial pigments of the aryl polyene type are structurally similar to the well-known carotenoids with respect to their polyene systems. Their biosynthetic gene cluster is widespread in taxonomically distant bacteria, and four classes of such pigments have been found. Here we report the structure elucidation of the aryl polyene/dialkylresorcinol hybrid pigments of Variovorax paradoxus B4 by HPLC-UV-MS, MALDI-MS and NMR. Furthermore, we show for the first time that this pigment class protects the bacterium fro… Show more

“…While the potential to form these linear precursors was observed, genes for squalene and lycopene cyclization were not identified. An aryl polyene biosynthetic gene cluster was identified in FLA‐8, which may produce a carotenoid‐like pigment possibly involved in photoprotection and scavenging of reactive oxygen species (Schöner et al ., ). An additional cluster in FLA‐8 contains genes for phytoene/squalene synthase, phytoene desaturase, lycopene cyclase domain‐containing protein and beta‐carotene hydroxylase (Supporting Information Fig.…”

“…kururiensis genomes. Aryl‐polyenes (APEs) are pigments similar to carotenoids and have been associated to bacterial protection against light‐induced damage and reactive oxygen species (ROS) (Cimermancic et al., ; Schoener et al., ). In all three genomes, the clusters comprise 40 kb and harbor approximately 45 genes, from which 35% of the genes show similarity with APE cluster from V .…”

“…According to AntiSMASH, the genome possesseses five secondary metabolite gene clusters with 14 putative gene clusters. Detected secondary metabolites include ectoin (osmolites protective substance) (Bernard et al 1983), resorcinal, arylopolyene (protects from reactive oxygen species) (Schöner Tim et al 2016), and terpene (ecological role), phosphonate (global phosphorus cycle) (Yu et al 2013) that supports the bacteria for surviving in harsh environment (Supplementary file 2).…”
