Ascorbic Acid Stabilization of Re-188- and I-131-Radiolabeled Peptides for Radiotherapy

Abstract: Re-188 labeled RC-160 [cyclic NH 2 -(D)-Phe-Cys-iyr-(D)-TrpLys-Val-Cys-Trp-NH 2 ] cyclic NH is a radiolabeled somatostatin analog which is being explored for its potential as a local/regionally administered radiotherapeutic agent targeting somatostatin-receptor-positive tumors. The stability of I88 Re-RC-160 towards radiolytic effects is a prerequisite for the success of such an approach. High radiation flux was found to result in radiolysis of the peptide, but addition of ascorbic acid to preparations of RC-1… Show more

“…Carrier added 188 Re(V)-DMSA was prepared according to the method of Bisundan et al [4] and modified by addition of ascorbic acid for protection against radiolysis [1,17]. Briefly, 1 ml of generator eluate was added to 10 mg DMSA (Aldrich), 0.5 mg (50 µl of a 10 mg/ml solution) ammonium perrhenate (Aldrich) and 5 mg ascorbic acid (Aldrich).…”

“…Some investigators have described the use of ascorbic acid, gentisic acid or a-tocopherol. [31][32][33] The most commonly used antioxidant is ascorbic acid. A combination of ascorbic acid and gentisic acid could also be used for example.…”
