Assessment of Catechol Induction and Glucuronidation in Rat Liver Microsomes

Abstract: ABSTRACT:Catechols are substances with a 1,2-dihydroxybenzene group from natural or synthetic origin. The aim of this study was to determine whether catechols (4-methylcatechol, 4-nitrocatechol, 2,3-dihydroxynaphthalene) and the antiparkinsonian drugs, entacapone and tolcapone, at doses 150 to 300 mg/kg/day, for 3 days, are able to enhance their own glucuronidation. The induction potency of catechols on rat liver UDP-glucuronosyltransferases (UGTs) was compared with that of a standard polychlorinated biphenyl … Show more

“…The glutathione conjugate formation may represent a mechanism to detoxify EGCG quinone. Such phenomena have been observed in previous studies with other flavonoids, dopamine, and catechol estrogens, which undergo peroxidase- or tyrosinase-catalyzed oxidation chemically, in primary cultured astrocytes and in the liver ( ). The thiol conjugates of catechol estrogens were detected in urine and kidney samples of hamsters ( , ), but no thiol conjugates of flavonoids were reported in vivo previously.…”

“…Glucuronidation has been described as being a major route for the conjugation of phenolic compounds such as acetaminophen ( 31 ) , quercetin ( 32 ) , PA and VA ( 33 ) , and other substances with a 1,2-dihydroxybenzene group ( 34 ) . However, sulfation is the primary conjugation route of the hydroxycinnamic acids CA, FA and DHCA ( 24 ) , and many other sulfated phenolics have been reported, including sulfates of PA and VA ( 35 ) .…”

“…Healthy and diseased mice were sacrificed using cardiac puncture either immediately or on day 12 after the GPI treatment, and the livers were collected and stored at −80°C until immediately before use. The MLM were prepared using previously reported protocols (17,18). The protein content of MLM was measured using the bidnchoninic add assay using a reported procedure (19).…”
