Aim:The aim of this study was to evaluate the relationship of scrotal circumference (SC) with plasma testosterone, seminal vesicles (SVs) weight, and its secretion as measurable indicators of fertility and also to sequence and establish phylogenetic relatedness of certain SV protein genes with other species as such integrated approach is lacking.Materials and Methods:Altogether, 59 apparently healthy male buffaloes sacrificed at slaughterhouse were selected (irrespective of breed) for measuring SC and collecting blood and paired SVs. The SC was measured at greater curvature using soft thread. In the present study, blood plasma testosterone, cholesterol, protein, and glucose in addition to SV fructose, citric acid and proteins in SV fluid were also estimated. The SV tissue was fixed in RNAlater for RNA extraction. Male buffaloes were categorized as per total SV weight into Group I (<5.0 g), Group II (5.0-7.84 g), and Group III (>8.0 g) and dentitions-I (≤18 months), II (18-24 months), and III (≥24 months) to assess the effect of weight and dentition age on SC, SV weight, and its certain secretions. Data were analyzed using linear model procedure including Tukey HSD test and Pearson’s correlation coefficient. Variance inflation and condition index were also used to assess multicollinearity.Results:Gross and histomorphological evaluation of SVs did not show any abnormality. Macronutrients (plasma protein, glucose, and cholesterol) showed non-significant (p>0.05) variation between groups. The SC and SV weight varied significantly (p<0.05) with a significant positive relationship with plasma testosterone, SV protein, fructose, and citric acid. In addition, testosterone concentration also showed increasing trend from Groups I to III but increased significantly (p<0.05) from Group II to III with positive and significant correlations with SV protein, fructose, and citric acid similar to SV weight and SC. Binders of sperm protein (BSP1, 3, and 5) genes (full length) were sequenced and established an evolutionary relationship which is lacking in buffalo.Conclusion:The present findings established a significant positive correlation of SC with that of other fertility parameters related to SVs weight and its secretions: Fructose, citric acid, and protein (inclusive of BSPs sequenced full length), and testosterone. Therefore, the present integrated approach along with certain semen quality attributes reflecting epididymis function could be used as a predictive fertility marker for grading and selection of breeding bulls and their progenies to develop outstanding bull mother farm.

Seminal vesicles were collected from 60 heads of Bulls which butchered in slaughter house (RPH) Yogyakarta. The aims of this study are knowing the relationship between body weight, age, fluids volume and concentration of prostaglandin F 2 α (PGF 2 α) in seminal vesicle of beef cattle. Those seminal vesicles were gathered from bulls which recorded its body weight and age then measured its seminal vesicles for weight, fluids volume and PGF2α levels. The PGF 2 α level was measured by Enzyme-Linked Immunosorbent Assay. Statistical analysis was performed using one way -analysis of varian, regression and correlation with P<0.05. Body weight of bulls showed positive correlation with the weight of seminal vesicle and its fluids volume. However, PGF 2 α levels were not correlated with the body weight of cattle. Weight of seminal vesicles also exhibited positive correlation with volume of vesicle fluids but no correlation with PGF 2 α levels. Based on the age of bulls, there were no correlation with the weight of seminal vesicles, seminal fluids volume and PGF 2 α levels. Those result indicated that the weight and fluids volume were affected by the body weight of bulls altough the PGF 2 α levels have a standard of development which seems affected by other factors such as concentration of androgen hormone.Keywords : cattle, body weight, age, seminal vesicle, PGF 2 α Abstrak Vesikula seminalis dikoleksi dari 60 ekor sapi yang dipotong di Rumah Potong Hewan Yogyakarta. Tujuan dari penelitian ini adalah mengetahui hubungan antara berat badan, umur, volume cairan dan konsentrasi prostaglandin F 2 α (PGF 2 α) dari vesikula seminalis sapi potong. Tingkat konsentrasi PGF 2 α diukur menggunakan Enzyme-Linked Immunosorbent Assay (ELISA) kit. Analisa statistik dilakukan menggunakan one way ANOVA, regresi dan korelasi dengan P<0.05. Berat badan sapi menunjukkan korelasi positif terhadap berat vesikula seminalis dan volume cairan yang dihasilkan. Konsentrasi PGF 2 α tidak berkorelasi terhadap berat sapi. Berat vesikula seminalis memiliki korelasi positif terhadap volume vesikula seminalis tetapi tidak berkorelasi dengan konsentrasi PGF 2 α. Berdasarkan pada umur sapi, parameter tersebut tidak berkorelasi dengan berat vesikula seminalis, volume cairan seminalis dan konsentrasi PGF 2 α. Hasil tersebut mengindikasikan bahwa tingkat konsentrasi PGF 2 α memiliki standar perkembangan tersendiri yang kemungkinan dipengaruhi oleh faktor lain seperti konsentrasi hormon androgen.Kata kunci: sapi potong, berat badan, umur, vesikula seminalis, PGF 2 α JSV 35 (1), Juni 2017

Aim:The experiment was conducted to study the total seminal plasma protein (TSPP) and heparin-binding proteins (HBPs) in relation to initial semen quality of buffalo bull. Materials and Methods:Semen from two Murrah buffalo bulls (bull no. 605 and 790) with mass motility of ≥3+ were used for the study and categorized into three groups (Group I-Mass motility 3+, Group II-Mass motility 4+ and Group III-Mass motility 5+). Seminal plasma from semen was separated by centrifugation. HBPs was isolated and purified from heparin-agarose affinity column by modified elution buffer. TSPP and isolated HBPs concentration was estimated by Lowry's method. The purified HBPs were resolved on Sodium dodecyl sulfate polyacrylamide gel electrophoresis to check the protein profile of two bulls. Conclusion: TSPP and HBPs shows variation in concentration with respect to initial semen quality. Furthermore, presence of fertility related 31 kDa HBPs in one of the bull may be an indication of high fertility of a bull. In future, in-vivo and in-vitro correlative study on larger basis is needed for the establishment of fertility-related HBPs in semen which might establish criteria for selection of buffalo bull with high fertility potential.
