Association between UBE2E2 variant rs7612463 and type 2 diabetes mellitus in a Chinese Han Population

Abstract: UBE2E2 encodes ubiquitin-conjugating enzyme E2E2, which plays an important role in the synthesis and secretion of insulin. Two previous studies indicated that SNPs in UBE2E2 were associated with risk for type 2 diabetes mellitus (T2DM) in the Japanese and Korean populations, respectively. We examined the association of one SNP in this gene, rs7612463, with the risk of T2DM in 1957 Han participants in northeastern China, using an SNPscan(TM) Kit. rs7612463 genotype was significantly associated with risk for T2D… Show more

“…Ubiquitin-conjugating enzyme E2E2 (UBE2E2) plays an important role in the synthesis and secretion of insulin. Mutations in UBE2E2 increase risk for type 2 diabetes [ 36 ]. 60S acidic ribosomal protein P0 is downregulated, suggesting a decrease in protein synthesis.…”

“…On chromosome 27, a QTL with strong evidence for MY (Table 3 ) corresponded to a single gene that aligned with mouse, rat and sheep sequences: UBE2E2 (Table 6 ). This gene is a good candidate for MY because it encodes a protein that plays an important role in the synthesis and secretion of insulin and may influence fat and protein content in milk [ 40 , 41 ].…”

“…Cheung et al suggested that MYC (MYC Proto-Oncogene) can serve as an important mediator of impaired insulin secretion and β-cell apoptosis 38 . Some researches have indicated that the SNPs in UBE2E2 (Ubiquitin Conjugating Enzyme E2) were associated with the development of type 2 diabetes mellitus 39,40 . Besides, MIB2 (Mindbomb E3 Ubiquitin Protein Ligase 2), ANAPC1 (Anaphase Promoting Complex Subunit 1) and TCEB1 (ELOC, Elongin C) are also involved in the process of ubiquitination which can affect the development of insulin resistance and metabolic syndrome [41][42][43] .…”
