Associations among central nervous system serotonergic function and neuroticism are moderated by gender

Abstract: Serotonergic dysregulation is associated with negative affect. Plasma prolactin responses to a tryptophan enhancement challenge are used as a measure of central nervous system serotonergic activity. We examined prolactin responses to a tryptophan challenge as they relate to the personality domains of Neuroticism, Extraversion, Openness, Agreeableness, and Conscientiousness. Participants were 67 volunteers. Regression models assessed peak prolactin response to intravenous tryptophan infusion as a predictor of N… Show more

“…Previous studies have shown that the serotonin system in men and women act differently in important ways (Biver et al 1996;Brummett et al 2008a;Costes et al 2005;Kornstein et al 2001;Nishizawa et al 1997;Williams et al 2003). Moreover, the 5HTTLPR genotype has different implications for personality traits and emotional well-being depending on sex (Brummett et al 2008c;Dragan and Oniszczenko 2006;Walderhaug et al 2007).…”

“…This neurobiological model of personality, based on the big five, links the metatrait stability (combination of neuroticism reversed, agreeableness, and conscientiousness) with variability in serotonergic function, and the metatrait plasticity (combination of extraversion and openness) with variability in dopaminergic function. Brummett et al (2008) found that high serotonergic function was linked with lower levels of neuroticism in healthy females. In a non-clinical sample, Manuck et al (1998) showed that in males, neuroticism and conscientiousness were related, negatively and positively, with high serotonergic function.…”

“…Few serotonergic challenge-studies have reported data regarding traits of positive emotionality (i.e., extraversion), most of which have not shown a link between extraversion and serotonergic responsivity using different substances (e.g., Manuck et al, 1998; Flory et al, 2004; Brummett et al, 2008; Kuepper et al, 2010). Indeed, Reist et al (1996) demonstrated a positive association of the cortisol response to paroxetin (indicating a diminished serotonergic responsivity) in healthy males.…”
