Background Cytokines were correlated with survival and disease progression in acute myeloid leukemia (AML). We aimed to evaluate the multivariate effect of TNF‐α rs361525, rs1800750, rs1800629, IL‐10 rs1800896, rs1800872, IL‐6 rs1800795, TGF‐β1 rs1800470, IFN‐γ rs2430561 single nucleotide polymorphisms (SNPs) on AML risk, the multivariate effect of SNPs on overall survival (OS) in AML and the association between the investigated SNPs and prognostic factors in AML. Methods All SNPs were genotyped in 226 adult AML cases and 406 healthy individuals. AML patients were investigated for FLT3 (ITD, D835), DNMT3A (R882), and NPM1 type A mutations. Results Univariate analysis revealed that age above 65 years had a negative influence on survival (P < .001). The presence of the rs1800750 variant genotype (P = .005) or FLT3‐ITD mutation (P = .009) in a cytogenetic high‐risk group (P = .003) negatively influenced OS. A negative association was observed between Eastern Cooperative Oncologic Group Scale status > 2, lactate dehydrogenase (LDH) level, platelet (PLT) count <40 000 cells/mm3, and OS. Multivariate Cox regression analysis showed that the presence of the rs1800750 variant genotype was a risk factor for death (P = .007), and that blast percentage, LDH level (≥600 IU/L), and cytogenetic high‐risk were independent significant predictors for death in AML (P = .04, corrected HR = 1.20; P = .022, corrected HR = 1.24; P = .021, corrected HR = 1.34, respectively). Conclusions Age above 65 years, PLT count, TNF‐α rs1800750 variant genotype, blast percentage, LDH level, and cytogenetic high‐risk may be used as independent risk factors to assess AML mortality.

The uptake of Fe(III) by living cells of unicellular green algae Dunaliella salina ions is investigated. The effects of operational conditions such as contact time, biomass concentration, initial pH, and initial Fe(III) concentration on iron uptake are studied. The time profile of the Fe(III) uptake by living cells of D. salina shows a time lag in the first 15 min, which is associated with the production of metal chelating agents. In addition, it is found that the optimum pH for maximal iron uptake is 8. At optimal conditions, the uptake is increasing gradually along with the increase of biomass concentration. Furthermore, the uptake increased with the rising of the initial metal ion concentration. Dry samples of iron free and iron containing biomass of D. salina are analyzed using Fourier‐transform infrared (FTIR) spectroscopy. Comparison of the obtained FTIR spectra revealed the presence of amino, carboxylic, hydroxylic, phosphate and sulphonate groups of the lipids, proteins and polysaccharides localized at the cell surface, which are involved in the process of Fe(III) acquisition.

Chronic inflammation can trigger events that would induce the malignant transformation of cells and carcinogenesis. Cytokines play a crucial role and can control the development and multiplication of cancerous cells. However, clinical data present controversy about the participation of these proteins in the establishment and development of cancer. Interleukin 10 (IL-10), a potent anti-inflammatory cytokine, has been the subject of multiple studies. Several studies have reported that IL-10 has pro-and antitumor effects. Elevated levels of IL-10 are associated with increased tumor growth with poor prognosis and drug resistance. However, this cytokine has both tumor-promoting and tumor-inhibiting properties. In vitro and in vivo studies report mechanisms by which IL-10 expression downregulates class I, which results in the control of the metastatic disease. IL-10 also inhibits tumorigenesis via downregulation of other cytokines. The variation observed could be the result of concentration ranges of this protein, genetic polymorphism, or both. The value obtained may serve as a biomarker indicative of tumor development and its prognosis.
