Astraodoric Acids A–D: New Lanostane Triterpenes from Edible Mushroom Astraeus odoratus and Their Anti-Mycobacterium tuberculosis H37Ra and Cytotoxic Activity

Abstract: Tuberculosis (TB) is one of the chronic infectious diseases caused by Mycobacterium tuberculosis that causes about 2-3 million deaths per year. Isoniazid and rifampicin are examples of first line drugs used for TB treatment; however, they are potentially hepatotoxic. More effective and safer drugs are urgently needed, especially from natural products. Basidiomycete mushrooms are known as important sources of pharmaceutically active metabolites including an anti-TB agent. In this work, the chemical constituents… Show more

“…We thus used the products from 95% ethanol extraction with sonication in this study. Although the active components in PFEC that exert the anti-tumor effects are not yet identified, the product is abundant in terpenoids, but not protein nor saccharides determined by a series of qualitative chemical reaction assays (data not shown), which is in line with other types of mushroom [25].…”

“…Compounds 1, 2, and 3 exerted signifi cant cytotoxic activity against MCF7 Brought to you by | Purdue University Libraries Authenticated Download Date | 5/30/15 5:27 AM cells with IC 50 values of 1.01, 1.54, and 2.15 μg/ml, respectively, and 3.49, 3.83, and 3.39 μg/ml against HCT116 cells, respectively. Previous investigations reported cytotoxic activity of lanostane triterpenes too (Ukiya and Akihisa, 2002;Akihisa et al, 2007;Monténégro et al, 2008;Zhou et al, 2008;Arpha et al, 2012). As compound 2 was found to be cytotoxic against the human cancer cell lines studied here, our results provide further support for the cytotoxic activity of lanostane triterpenes.…”

“…The most abundant chemical constituents found so far in these genera are triterpenoids, which exhibit various biological activities such as, antifungal, antibacterial, antituberculosis and cytotoxicity against cancer cell lines (Kope et al, 1991;Stanikunaite et al, 2008;Tapan et al, 2012). Our studies into A. odoratus also isolated four new lanostane triterpenoid astraodoric acids A-D with astraodoric acids A and B exhibiting antituberculosis and anticancer activities (Arpha et al, 2012). A modified astraodorol also exhibited the antimalarial activity that had been reported by Nasomjai et al, 2014.…”

“…A modified astraodorol also exhibited the antimalarial activity that had been reported by Nasomjai et al, 2014. Further research of the bioactive chemical constituents in these genera has led to the isolation of two lanostane triterpenoids derivatives (2-3) and six known compounds including astraodorol (Arpha et al, 2012), artabotryol B (Gupta et al, 2010), artabotryol C1 (Gupta et al, 2010), 6-dehydrocerevisterol (Lee et al, 2010), ergosterol (Arpha et al, 2012) and hypaphorine (Arpha et al, 2012) from the edible mushroom A. asiaticus, which are described below.…”
