ATP Hydrolysis Is Essential for the Function of the Uup ATP-binding Cassette ATPase in Precise Excision of Transposons

Abstract: In Escherichia coli K-12, the RecA-and transposase-independent precise excision of transposons is thought to be mediated by the slippage of the DNA polymerase between the two short direct repeats that flank the transposon. Inactivation of the uup gene, encoding an ATP-binding cassette (ABC) ATPase, led to an important increase in the frequency of precise excision of transposons Tn10 and Tn5 and a defective growth of bacteriophage Mu. To provide insight into the mechanism of Uup in transposon excision, we purif… Show more

“…revealed neither recombination nor excision footprints but only the initial target TA, which highlighted the mechanism of perfect excision described in the PiggyBac element Grossman et al, 2000), the Mu-like transposon Hop (Chalvet et al, 2003), in Tn10 and Tn5 bacterial transposons (Murat et al, 2006) and in Restless elements after integration in Neurospora crassa (Windhofer et al, 2002). Although no new insertion could be demonstrated in O. novo-ulmi, we believe that a combination of introgression and abiotic stresses may act in favor of the invasion of TEs from one species to a closely related one.…”

“…However, as tested here, vanadate is clearly not an inhibitor of the SgA resistance mechanism in staphylococci. Insensitivity to vanadate might be a general feature of the ART family of ABC systems that comprise homologues to both Vga(A) and Uup (15), an E. coli protein structurally and phylogenetically related to Vga(A) but not involved in drug resistance (37). A specific conformation of the ATP-binding sites might prevent the replacement of P i by vanadate in these ABC ATPases.…”
