Atypical B cells are part of an alternative lineage of B cells that participates in responses to vaccination and infection in humans

Abstract: Highlights d Single-cell RNA-seq reveals two distinct B cell lineages d An alternative lineage contains CXCR3 + and atypical B cells d Alternative B cells are primed after primary vaccination and respond to boosters d Alternative B cells adopt a more atypical phenotype following repeated antigen exposure

“…As a result of the role of eATP as mediator of systemic inflammation, 54 we propose that elevated levels of CD27 − IgD − B cells in circulation are, at least partially, a reflection of a vigorous strong immune reaction with no detriment for the development of efficient antiviral immunity. This is in line with recent findings that CD27 − IgD − B cells can be detected during normal immune reactions 55 . Our findings have implications for both our basic understanding of human B‐cell responses to virus infections and reveal that ATP is released during acute hantavirus infection.…”

“…We used publicly available paired PBMC and FNA single cell sequencing to demonstrate FCRL5 + B cells are circulating progeny from the GC reaction. Recently, an FCRL5 + population has been described after malaria vaccination in naïve volunteers (Sutton et al, 2021), and our data supports this observation that FCRL5 + B memory cells can be part of a ‘typical’ immune response. In this study, atypical memory B cells from older persons showed evidence of SHM, but less evidence of positive selection, indicating impaired GC function in older people.…”

“…Total isotype-switched B cells from two malaria exposed adults and two unexposed adults were also analyzed by scRNA-seq, and the results provided evidence of heterogeneity within ABCs (three clusters), MBCs (three clusters), and activated B cells (two clusters) similar to our description here of new heterogeneities in all B cell subpopulations including naïve B cells that were not included in the Sutton et al . ( 45 ) analyses. A pseudotime trajectory was also provided, showing that MBCs and ABCs independently differentiated from naïve B cells.…”
