Augmented Contributions of Voltage-Gated Ca2 Channels to Contractile Responses in Spontaneously Hypertensive Rat Mesenteric Arteries

Abstract: The observation that organic Ca2+ channel blockers are more effective in lowering blood pressure and peripheral resistance in hypertensive compared to normotensive subjects suggests that there is a greater contribution from voltage-gated Ca2+ channels (CaL) to vascular force maintenance in hypertensive arteries. This study tests this hypothesis by comparing the effects of Bay k 8644 and nisoldipine on basal force development, contractile responses to norepinephrine and serotonin, and Ca2+ currents (ICa) in mes… Show more

“…This is in agreement with Matsuda et al [28]in rat mesenteric arteries and with Tosun et al [29]in the rat aorta, but in contrast to Dorn and Becker [30]who did not find evidence for TxA2-mediated Ca 2+ entry in aortic smooth muscle cells. In the latter two studies, experiments were conducted on either rat aortic rings in isometric conditions [29]or isolated aortic smooth muscle cells [30], which represent two very different experimental conditions in a large compliance artery.…”

“…26 VSM isolated from hypertensive SHRs shows augmented contraction in response to stimulation of voltage-dependent Ca 2ϩ channels by Bay K 8644. 27 In addition, blood vessels from SHRs exhibit upregulation of the L type of Ca 2ϩ channel expression. 28 Different from adult hypertensive SHR, vascular PI3K-C2␣ activity in young prehypertensive SHRs, in which alterations in Ca 2ϩ channel activity are not detected, 26 was not elevated compared with normotensive WKY rats (Figure 3).…”

“…These changes result in an increased contribution of L-type Ca 2ϩ channels to agonist-mediated responses in hypertensive ASM. 1,2,39 This in turn increases the effectiveness of L-type Ca 2ϩ blockers in decreasing vascular tone and blood pressure in hypertensives. 40 K v channels are an important determinant of resting membrane potential in ASM.…”
