Autoantibody reactive with three classes of RNA polymerases in sera from patients with systemic sclerosis.

Abstract: We have identified a novel autoantibody reactive with all three classes of RNA polymerases, well-characterized nuclear enzymes, in sera from patients with systemic sclerosis (SSc).

“…The followup period was <5 years in 85 patients, [5][6][7][8][9][10] years in 53 patients, and > 10 years in 75 patients. During the period of observation, 99 patients received corticosteroids (mean maximum dosage 33 mg of prednisone equivalent daily), 23 received D-penicillamine, and 16 received cytotoxic drugs (11 took azathioprine, 3 methotrexate, and 2 mizoribine).…”

“…We performed indirect immunofluorescence tests using commercially prepared slides of monolayer HEp-2 cells (MBL, Nagano, Japan) as substrate (lo), double-immunodiffusion tests using calf thymus extracts as antigen source ( 3 3 , and immunoprecipitation assays using 32P-orthophosphate-labeled (15) (10). Anti-top0 I was considered positive if the serum antibody produced a precipitin line identical to that produced by the reference serum on double immunodiffusion, or if it immunoprecipitated a protein with a molecular weight of -100 kd from "S-methionine-labeled cell extract, inhibited top0 I enzymatic activity in vitro using commercially prepared calf thymus top0 I (Takara, Kyoto, Japan) according to the method of Goto et a1 (37), and reacted with any top0 I fusion protein including B cell epitope(s) on immunoblots, as described elsewhere (35).…”

“…These SSc-associated antibodies are useful in the diagnosis of SSc and classification of patients, and thus are widely used in clinical settings (1,2). In addition, sera from a subset of SSc patients recognize 1 or more subtypes of RNA polymerase (RNAP) (2)(3)(4). Autoantibodies to RNAP I and RNAP III always coexist, and this pattern of antibody response is highly specific for SSc (2)(3)(4).…”

“…tween hnRNPs and RNA polymerase, since some hnRNP proteins assemble on RNA polymerase I1 transcripts as soon as they are synthesized (1,35) and autoantibodies to RNA polymerases occur specifically in SSc (36,37). We believe that studies on the association of anti-hnRNP I with different SSc-associated autoantibodies might provide new insights on co-localization and function of the hnRNP I protein.…”
