Automated Broadband Phase Correction of Fourier Transform Ion Cyclotron Resonance Mass Spectra

Abstract: It has been known for 35 years that phase correction of FTICR data can in principle produce an absorption-mode spectrum with mass resolving power as much as a factor of 2 higher than conventional magnitude-mode display, an improvement otherwise requiring a (much more expensive) increase in magnetic field strength. However, temporally dispersed excitation followed by time-delayed detection results in steep quadratic variation of signal phase with frequency. Here, we present a robust, rapid, automated method to … Show more

“…The molecular mass of each peak was determined by deconvolution of multiply charged ion species and then compared with the calculated masses of all histone H2B sequence variants (Table I). All raw ECD product ion mass spectra were phase-corrected and analyzed by our in-house software in a variant-specific man-ner to determine each proteoform (55,56). Briefly, a library of all known H2B sequence variants from Uniprot was imported into our software for identification and characterization.…”

“…For FT-ICR, it was recently demonstrated that, because of linear frequency sweep excitation, Φ(ω) is a quadratic function [32,33]. Recovery of the absorption-mode spectrum from the complex spectrum requires the correct phase angle function, Φ(ω); see Eq.…”

“…Individual time-domain transients (80 -200; 6.1 sec each) for each spectrum were signal averaged, apodized with a full-Hanning (magnitude mode) or half-Hanning (absorption mode) weight function, and zero-filled once, prior to fast Fourier transformation. [41,42] The time-signal did not dampen to zero during the acquisition period. Due to increased complexity at higher m/z, a broadband phase correction was applied to each mass spectrum to produce absorption-mode display with increased mass resolution.…”
