Avian adenovirus vector CELO-TK displays anticancer activity in human cancer cells and suppresses established murine melanoma tumors

Abstract: Avian adenovirus CELO is a novel adenovirus vector system with the advantages of efficient production, high virion stability, and the absence of crossreactivity with Ad5-neutralizing antibodies. In this study, we evaluated the anticancer efficacy of a CELO vector encoding the herpes simplex virus type 1 thymidine kinase, a prodrug-activating therapeutic gene. Vectors carrying the gene for HSV-tk or EGFP under the control of the HCMV promoter in place of the ''nonessential'' region of the CELO genome were const… Show more

“…39 In addition to this cytoskeleton transport of the virus, a putative nuclear localization signal (MAPTKRKGERKD) in the N-terminal region of VP1 may mediate nuclear translocation through nuclear pore. 36 The virus would seem then to uncoat, which exposes the encapsidated DNA to the nucleus allowing gene expression. The intracellular trafficking pathway of polyomavirus is different from that of retrovirus vectors.…”

“…The mice were intraperitoneally injected with GCV (100 mg kg -1 ) daily after the first IV injection of the tk-VLP or PBS. Tumor volume was determined using the equation: (a 2  b  P)/6 as described earlier, 36 in which 'a' is the tumor width and 'b' the tumor length. Measurements were performed twice a week with a caliper.…”

“…Human adenoviruses (HAdVs) and other mammalian adenoviruses are used both as oncolytic viruses (10)(11)(12) and vaccine vectors (13,14). FAdVs are also suitable vectors; for example, FAdV-1-and FAdV-8-based recombinant viruses have induced protective immune responses against infectious bursal disease virus and infectious bronchitis virus, respectively (15,16).…”

“…The E3 region in these vectors is identical to the E3 region in KD3 allowing for overexpression of ADP in the absence of immunomodulatory E3 genes. 32 The TetON-EGFP and TetON-SEAP vectors were constructed for visualization 33 and quantification 30 of transgene expression, respectively.…”
