Avocado Cellulase Extraction and Purification

Abstract: Optimum conditions for the extraction and purification of avocado cellulase have been established. The best quantiative recovery of the cellulase from avocado tissue was obtained with a 0.04M acetate buffer, pH 5.0, containing 0.4M NaCl and 0.25% (v/v) Triton X-100. The isoelectric point of the enzyme was 4.7 and the molecular weight, based on SDS gel electrophoresis, was 49,000. The enzyme was purified on a cellulose column to a single protein band on SDS gel electrophoresis. Rabbit antibody produced to the c… Show more

“…Whereas this measure does not account for the tertiary structure of the 695 mature protein, it does reflect their relative acidic or basic nature. The deduced pI for TPP18 of 9.2 is very different from that of avocado protein which is predicted to be 5.3 and has been shown to actually be 4.7 [3]. The apparent basic nature of TPP18 is more similar to that of the bean endo-fl-1,4-glucanase (BAC) which has predicted and measured pI values of 7.8 and 9.5, respectively [30] and of tomCell and tomCel2 which have predicted pls of 7.3 and 8.2, respectively [50].…”

“…Cellulose anity chromatography has been used to purify EGs from a range of tissues, including the fruits of avocado (Awad and Lewis 1980), pepper (Ferrarese et al 1995) and apple (Abeles and Biles 1991), as well as bean leaf (Durbin and Lewis 1988), sweet pea anthers (Sexton et al 1990), and elder leaf (Webb et al 1993).
