Axon position within the corpus callosum determines contralateral cortical projection

Abstract: How developing axons in the corpus callosum (CC) achieve their homotopic projection to the contralateral cortex remains unclear. We found that axonal position within the CC plays a critical role in this projection. Labeling of nearby callosal axons in mice showed that callosal axons were segregated in an orderly fashion, with those from more medial cerebral cortex located more dorsally and subsequently projecting to more medial contralateral cortical regions. The normal axonal order within the CC was grossly d… Show more

“…De-fasciculation of cingulate callosal axons has also been reported in mice with specific deletion of Nrp1 in the primary motor cortex (Zhou et al, 2013), very similar to our observations in Emx1 −/− /C57Bl/6 mice. We therefore hypothesized that NRP1/SEMA signaling might be disrupted in the Emx1 −/− /C57Bl/6 mice.…”

“…NRP1/SEMA3C signaling is required for the midline crossing of callosal axons Axon targeting and de-fasciculation defects that are due to a loss of NRP1/SEMA signaling are well documented for neocortical callosal axons in vivo (Gu et al, 2003;Hatanaka et al, 2009;Zhao et al, 2011;Zylbersztejn et al, 2012;Zhou et al, 2013). Together with recent studies (Niquille et al, 2009;Piper et al, 2009), our findings demonstrate that NRP1/SEMA signaling regulates similar events during the development of cingulate callosal axons.…”
