In alkaline conditions, Listeria monocytogenes cells develop higher proportions of branched-chain fatty acids (FAs), including more anteiso forms. In acid conditions, the opposite occurs. Reduced growth of pH-sensitive mutants at adverse pH (5.0/9.0) was alleviated by the addition of 2-methylbutyrate (an anteiso-FA precursor), suggesting that anteiso-FAs are important in adaptation to adverse pH. The balance between anteiso-and iso-FAs may be more important than changes in the amounts and/or degrees of saturation of FAs in pH adaptation.Listeria monocytogenes can grow under a wide range of pH stress, i.e., 4.1 to 9.0 (38, 46), increasing its abilities to persist during food processing and attempts to decontaminate food-processing environments (15,(42)(43)(44)(45). It also has particularly impressive capacities to modulate its membrane lipids to maintain membrane fluidity and transport functions (10,40,41) in response to temperature (1), salt (7), and CO 2 /anaerobiotic (21) stress. Such capacities have been suggested to be related to its atypically high iso and anteiso, odd-numbered, branched-chain fatty acid (BCFA) content (1,20) and its ability to modulate the overall content and proportions of BCFAs, straight-chain FAs (SCFAs), and unsaturated FAs (22,23). For example, reductions in environmental temperatures lead to increases in the amount of ai15:0 present in L. monocytogenes cell membranes, while increases in environmental temperatures lead to reductions in the amounts of ai15:0 and other BCFAs present in membranes (1,14,32).Changes in FA profile have been associated with pH adaptation in Streptococcus mutans (16-18), Escherichia coli (5, 48), and Salmonella (24), Pseudomonas (31), and Bacillus species (23). However, little is known about pH stress-associated modulation of FAs in L. monocytogenes (21), the wider role of FA modulation in its responses to nonthermal stresses, or the cross-protection mechanisms which operate in this hardy pathogen (19,27,35,44).The aims of this study were to investigate the modulation of the FA profile of L. monocytogenes membranes in response to changes in environmental pH, investigate the effects of an exogenous BCFA precursor on the pH stress response of BCFA-deficient mutants (1, 49), and examine possible links between the prevalence of anteiso-BCFAs and the adaptation mechanism(s) of L. monocytogenes under adverse pH conditions. Modified brain heart infusion broth (MBHIB; Difco Laboratories, Sparks, MD), suitable for adverse-pH studies, was prepared to pH 5.0, 5.5, and 6.0 in 2 M disodium phosphate Washed cells from mid-exponential-phase cultures of L. monocytogenes 10403S, an isogenic sigB null mutant (3), and isogenic non-BCFA-producing cld-1 and cld-2 mutants (1, 49) were inoculated into preheated (30°C) 100-ml volumes of the buffered MBHIB and grown (30°C/200 rpm) to an optical density at 600 nm of 0.5 to 0.6. An acidophilic, disulfide-oxidizing, mesophilic, aerobic bacterium was isolated from wastewater sludge. The new organism is a gram-positive sporulated rod. It can use elemental sulfur and pyrite as sole energy sources and grows on organic substrates such as glutamate and glucose. It also grows on the following organic sulfur substrates: oxidized and reduced glutathione, cysteine, cystine, and dithio(bis) benzothiazole and clearly shows a preference for disulfide bond-containing substrates. The optimal pH of growth is between 1.5 and 2.5, depending on the substrate used, and the growth temperature range varies from 4 to 40°C, with an optimal value at 35°C. The G+C chromosomal DNA content was measured at 53 A 1 mol%. Phylogenetic analysis of 16s genes coding for rRNA sequences places the new isolate in the genus Sulfobacillus. In addition, unique phenotypic and physiologic characteristics and DNA homology values assign the isolate to a new species in the genus. Therefore, this new isolate has been named Sulfobacillus disulJidooxidans and has been assigned ATCC number 51911.Many facultatively heterotrophic bacteria are known for their ability to grow under highly acidic conditions. Some of these within the genera Sulfolobus (5) and Acidianus (31) are extremely thermophilic archaebacteria. They obtain their energy by the oxidation or reduction of elemental sulfur or from organic substrates. Others are mesophilic gram-negative bacteria belonging to the genus Acidiphilium (2, 18) and are able to use organic substrates at pH 3.0 or belong to the genus Thiobacillus, such as Thiobacillus acidophilus (17), which is also known for its ability to grow with elemental sulfur or glucose at pH 2.5.However, the only acidophilic, facultatively autotrophic, gram-positive, spore-forming bacillus so far characterized is Sulfobacillus thermosu&dooxidans (16), together with two subspecies ("themzotolerans" and "asporogenes") that do not produce spores (19). Golovacheva and Karavaiko (16) first described the genus Sulfobacillus in 1978 as gram-positive, sporeforming bacteria growing on elemental sulfur, ferrous iron, and other metal sulfides. The rod-shaped cells were described as fairly pleomorphic. The spores were spherical or slightly oval, terminal, subterminal, or paracentral. S. therrnosulfdooxidans is a typical acidophilic, thermotolerant, and facultatively autotrophic bacterium. Since then, four isolates which are gramvariable, thermotolerant acidophiles have been recovered from mines: ALV (27), BC (27), C-MT1(15), and TH3 (4), and have been described as belonging presumably to this genus. Bacteria of the genus Alicyiclobacillus spp. form spores and develop in acid media, leading to the spoilage of citrus juices. Brazil is the largest exporter of orange juice concentrate, and yet, it has been extensively studied due to changes in taste and smell. Several investigations have reported different culture media used to detect and enumerate Alicyiclobacillus spp. The objective of the present study was to evaluate the recovery of Alicyiclobacillus spp. spores grown in ALI, BAT, K agar and YSG media using the methodology suggested by ABECitrus. Five inocula were used, two from reference strains and three from pasteurized concentrated orange juice. Cell recovery after the enrichment in reconstituted orange juice was also analyzed. An initial population of 6 log CFU/mL was inoculated. ALI, BAT and YSG media were able to recover the initial population of all different inocula, with no significant differences between the results. When compared to BAT, however, the preparation of ALI and YSG media was simpler and had more advantages. The recovery with K agar medium was lower than the other media for all the tested inocula, with significant differences found for Alicyclobacillus acidocaldarius 0298T (3.66 log CFU/mL) and Alicyclobacillus pomorum-like CBMAI 0278 (4.11 log CFU/mL).
